Objective:By contrasting three different genotypes, GG、GA、AA, all of which locate inthe CyclinA2gene rs769236locus,and analysising the association between the sitegene polymorphisms and colon cancer susceptibility, we could research the differenceof frequency between colorectal cancer patients and healthy people from Chinese Hanpopulation. Also,this study provides us new theory and method for the earlydiagnosis、therapy and posttreatment-estimateing of colorectal tumor.Method:We Collected150examples of peripheral blood from patients who werediagnosed colorectal cancer and150ones from healthy people, both of these peopleare independent individuals from each other. Then we extracted genomic DNA fromthose examples. At the same time, a database is set up by collecting clinicalpathological data and health questionnaire in allusion to the patients who weredesigned into two groups including experimental and control groups. The rs769236polymorphism of CyclinA2were analyzed by PCR-restriction fragment lengthpolymorphism(PCR-RFLP)in CRC cases and healthy controls. The associationbetween CRC risk and the SNP(rs769236) was estimated by an unconditional logisticregression model.Result:Compared to GG Wild genotype, Variant genotypes(GA+AA) carriers had asignificantly increased risk of CRC[adjusted odds ratio (OR)=1.92,95%confidenceinterval(CI)=1.10-3.36].In the stratified analysis,rs769236genetic effect was morepronounced in the people with family history of cancer [adjusted odds ratio (OR)=2.01,95%confidence interval(CI)=1.10-3.65,P=0.023]Conclusion:The current results suggested that the SNP(rs769236) of CyclinA2is associatedsignificantly with increased risk of CRC, the Variant genotypes(GA+AA) are theindependent risk factors of CRC in Chinese Han population.
