BackgroundIn the various types of malignant tumors of the digestive system, gastriccancer is a common one, at any age. The incidence increasing was graduallygrowing trend with age. Its main causes have: diet, genetic factors, and stomachdiseases. Rhein is from the rhubarb, which belong to the anthraquinonecompounds. According to the reports, Rhein increasing the Ca2+contentintracellular, which reduce mitochondrial membrane potential, with activation ofthe mitochondrial signaling pathway to promote apoptosis[1,2]. In the body,theresearch of Rhein is little, the mainly cause of which the Rhein is insoluble inaqueous solution. The anthraquinone compounds have water-insoluble in common.In this study, based on lysine change Rhein water-soluble, we have the RHL bychange the structural of Rhein, which water-soluble far greater than the RheinIn the1996,Parker F the first cloned G3BP gene from living Chinese hamsterlung fibroblasts[3]. It is located in the cytoplasm, which bind to RasGAP SH3domain and participate in Ras downstream signaling pathways. The relevantstudies, in many cell signaling pathways, the function of the RasGAP SH3domains is indispensable, but also the Ras pathway regulate Rho-mediatedapoptosis, and the cytoskeleton reconstruction[4][5].The relationship of the G3BP with tumor, there are a lot of research andfound. The appearent of the G3BP N end in particular the monoclonal antibodywhich make it like a milestone in antitumor therapy. With the continuousdevelopment of society and technology, there will be more in-depth explorationand research on the role and function of G3BP.By MTT, Western blotting analysis, RT-PCR analysis of apoptosis analysismethod of the AnnexinV2FITC/PI method,the research explore the the RHL ongastric cancer cell line AGS cell proliferation and apoptosis mechanisms andG3BP1signal the role of the pathway in the mechanism Objective:By the RHL,research proliferation, apoptosis mechanism and G3BP1signaling pathway in the mechanism of gastric cancer cell line AGS cells.Methods:With the MTT cell proliferation; protein apoptotic Westernblot; applicationof RT-PCR detection of the G3BP1mRNA expression level; apoptosis by flowcytometry.Results:RHL can inhibit gastric cancer cell line AGS cell proliferation and induceapoptosis, with increasing drug concentration, the apoptotic rate increasedgradually;RHL suppression G3BP1protein by Western blotting; RT-PCR resultsshow that the RHL can inhibit the transcriptional level G3BP1mRNA, therebyinhibit its protein expression.Conclusion:1、RHL inhibit the proliferation of gastric cancer cell, which show significantconcentration-dependent manner;2、RHL inhibit G3BP-1gene expression and induce gastric cancer cellapoptosis by G3BP-1pathway.
