Cynanchum bungei Decne. originates from the asclepiadaceous plant. C. bungeiDecne. is a perennial herb, scatters broadly in the region of Shandong, Hebei, Gansu ofChina,and is especially famous in Taian of Shandong. As our country’s special tonictraditional Chinese medicine, C. bungei Decne medicinal materials used to circulate onthe market are mainly crude drug, the traditional Chinese medicine and health careproducts in its other dosage forms are still to be developed. Up to now, It relied on wildresource for medicinal source, factors such as the long growth period, the low yield, theexcessive excavation, and so on, led to a sharp drop in wild resources, there were alsomassively the radix Cynanchum auriculatum Royle ex Wight from the same genus butdifferent species on the market, they were different in chemical composition, so theyshould not mix up. Lack of C. bungei Decne medicinal source had a serious impact onneeds of clinical medications and its developing and using.To develop and increase C. bungei Decne medicinal source, a specie of high-qualityand high-yield production had been selected through domestication and systematicselection. Its name is Taibai No.1. In this paper, the observation and the research aboutTaibai No.1biological characteristics were constructed, at the same time medicinalcharacteristics, medicinal quality and C. bungei Decne. molecular biological featureswere studied to provide a basis for planting and populating Taibai No.1. Results showedthat:1. Taibai No.1biological characteristics Objective: Make sure the breed character of the new C. bungei Decne variety steadyand the yield and quality of its medicinal characteristics wonderful, and then provide thebasis for agricultural industrialization of Taibai No.1.Methods: Select draughty drafty and broad field as the experimental field, sow theseeds by early April, rarefy young plant properly, steady young plant, and make sureappropriate field management. And then observe and compare their biologicalcharacteristics between Taibai No.1and the traditional C. bungei Decne spawn, whichmainly includes: seed analysis, botany features, growth and development law, harvest,and so on.Results and conclusions: The fruit of Taibai No.1represented the advantages thatthe fruit was big and seeds were numerous; The seeds of Taibai No.1were also greatlyfull; the plants of Taibai No.1represented that plants growth was straight, they werevigous and dark-green in leaf colour, the root had the advantage of good shape with lessfibril and more axillary branching, the yield per plant was higher, for these reasons, itsmain agronomic characteristics were better than the traditional spawn.2. Determine contents of the main active ingredients in different C. bungei DecnecultivarsObjective: Determine contents of baishouwubenzophenone and baishouwuyisu indifferent C. bungei Decne cultivars, provide the basis for the quality evaluation of C.bungei Decne medicinal materials.Methods: The samples were extracted by methyl alcohol in acelonnitrile-waterbefore detected by HPLC-UV. The analytical column was C18, the mobilepase wasacetonitrile-water(30:70), the flow rate was0.8mL/min, the detection wave length was280nm, injection volume was10μL.Results and conclusions: Good linearities were observed in the range of0.1~200μg/mL, with correlation coefficients were0.9992,0.9984. Contents of the two mainactive ingredients in the samples were stable within48hours; baishouwubenzophenoneRSD was5.09%, baishouwuyisu RSD was2.96%. The recoveries of the two compoundswere all between94.79%and101.12%, and the relative standard derivations were both below8%. The method conformed to the requirements of the instrument analysis.Contents of baishouwubenzophenone and baishouwuyisu in the Taiwu No.1medicinalmaterials were respectively0.755,0.017mg/g, were both higher than that in thetraditional spawn.3. Identify different C. bungei Decne varieties by using ITS and psbA-trnH intergenicregionsObjective: Make the differences of ITS and psbA-trnH intergenic regions amongdifferent Cynanchum bungei Decne varieties clear, and provide referential basis for DNAbacording and molecule biological study of C. bungei Decne.Methods: Using two universal pairs of primers (ITS1-ITS4, psbAf-trnHr), ITS andpsbA-trnH were amplified by PCR, amplification products were cloned and sequenceafter recovery and purification, and then compare and analyze variation characteristicswith MEGA, DNAMAN software and so on.Results and conclusions: Different C. bungei Decne varieties were same in thepsbA-trnH sequences. Their ITS sequences may be different anywhere from slightly toradically, the five samples from the Taibai No.1plant base form an independent branch.It illustrated that the five samples from the Taibai No.1plant base may be the samevariety, Taibai No.1was equipped with relative genetic stability. The ITS sequences andpsbA-trnH sequences made gene banks richer, and laid a foundation for the molecularidentification of C. bungei Decne and Taiwu No.1.
