Malignancy is a disease that pose a serious threat to the human’s health, as the effectiveness of surgery for extensive transfer and recurrence in terms of advanced malignancies, so that people’s attention focus on the target therapy for tumor. The literature suggests that exercise has a certain effect on the tumor treatment, so the movement was intruduced to the treatment about malignant tumors, and confirmed that it has a certain impact on the targeted treatment of the tumor, but it is unclear about the way of its influenced. In this study, we will observe the the impact of the movement about the tumor-targeting drug delivery system through C57BL/6mouse model that suffering from Hepal-6tumor; and it’s influence about the therapeutic effect of nanoparticles upload hydrochloride Mitoxantrone of mPEG-PLGA-PLL-cRGD, and investigate the possible mechanism that movement affect the treatment effect of the tumor targeted drug delivery.Experimental methods:1. Mitoxantrone hydrochloride as a model drug, to mPEG-PLGA-PLL-cRGD as the carrier, the use of double emulsion preparation of Mitoxantrone hydrochloride contained in mPEG-PLGA-PLL-cRGD is satisfied that the rice through quality evaluation of nanoparticles and Hepal-6cells in vitro MTT test, proved Mitoxantrone hydrochloride contained in mPEG-PLGA-PLL-cRGD receiving the rice preparation and on growth of Hepal-6cells in vitro。2.The6to8weeks male C57BL/6small rats was select to estabilish tumor model that contains Hepal-6,78male C57BL/6small rats are devided averagely into seven groups by random: control groups; sport groups; Mitoxantrone groups; Nanoparticles groups; sport joint Mitoxantrone groups; sport joint mitoxantrone nanoparticle that contains Hydrochloric acid groups. During the experiment, The abdominal cavity of mice was inject saline that belongs control groups and sport groups; The Mitoxantrone groups and sport joint Mitoxantrone groups was inject fee mitoxantrone hydrochloride solution; The Nanoparticles groups sport joint mitoxantrone nanoparticle that contains Hydrochloric acid groups was injected mitoxantrone hydrochloride nanoparticles solution that upload Hydrochloric acid. After the solution was injected into the rat’s body, The exercise stress A certain intensity exercise stressand last14day excersise was given related groups. Between the experiment, to measure the body weight of mice and tumor volume every other day. And measure the NO content of serum and aortic VEGF content levels after the exercise.Experimental results: 1. Confirmed that when Upload mitoxantrone hydrochloride mPEG-PLGA-PLL-cRGD nanoparticles,it having cytotoxic for the Hepal-6cells, while the blank material MPEG-PLGA-PLL-cRGD nanoparticles has no effect on is apoptosis.2. Movement combined with tumor targeted drug delivery system an significantly inhibit the growth of tumors of tumor-bearing mice; after two weeks exercise, NPS group and SP+NPS group hae a better inhibit rate about the tutor; The SP group、DHAQ group、SP+DHAQ group’s inhibitory effect foe the tumor is poor; About the SP group, SP+DHAQ group, SP+NPS group, the serum NO and aortic VEGF levels were significantly higher than the Control group; The concentration of DHAQ SP+DHAQ group the SP+NPS group is higher than DHAQ group and the NPS group. Note exercise can enhance the inhibitory effect of drug-loaded nanoparticles for drug delivery system.Conclusion:1. Exercise can be used as an auxiliary intervention to Improve the therapeutic effect of the active peptide the modified upload hydrochloric mitoxantrone nanoparticles.2. The possible mechanisms that Appropriate exercise to enhance the therapeutic effect of the drug tumor targeted drug delivery is:movement promote the generation of NO and VEGF, maybe enhancing the combination of the RGD peptide and the integrin αaβ3growth and through to promote apoptosis of tumor cells to inhibit the growth of tumor.
