objective: This study was conducted to determine theexpression of podoplanin and Bmi-1in nasopharyngeal carcinomapatients.All patients were treated by primary radio(chemo)therapy.Methods and Materials: The expression of podoplanin in HNE-1nasopharyngeal carcinoma cell lines were determined byimmunohistochemistry.Collected97cases of nasopharyngeal carcinomapatients who underwent primarily radical radio(chemo)therapy between2008to2009, The expression of podoplanin and Bmi-1in primary lesionof97nasopharyngeal carcinoma was evaluated byimmunohistochemistry, their correlation with patients’clinical-pathological data and the correlation between them wereanalysed.The correlation was evaluated by chi-squre or Fisher’s exact test.Followed up overall survival rates、local control rates、distantmetastasis free survival rates、disease special survival rates for threeyears of the97patients,and survival rates were calculated byKaplan-meier test,significance test using log-rank test.Prognostic factorsanalysis before treatment was used sigle prognostic factor analysis andCOX proportional harzad model. Results: The expression rate of PDPN in the dense parts of HNE-1nasopharyngeal carcinoma cell line was high, but in cancer cells of97cases of nasopharyngeal carcinoma patients it was low, The expressionrate of PDPN in tumor specimens was only4.1%;but it was high in lymphendothelial cells and in mucosal basal cells. Bmi-1was expressed inmost tumor cells and lymphocytes, the positive rate was57.1%, the highexpression rate was22.7%in nasopharyngeal carcinoma. PDPN, Bmi-1were diffusely expressed in tumor cells, none of them was expressed ininvasive edge of tumor. Chi-square test showed the expression of PDPNin nasopharyngeal carcinoma tumor cells had no correlation with tumorrecurrence、 distant metastasis、cervical lymph node metastasis、retropharyngeal lymph node metastasis、 T stage、clinical stage、Nstage, and had no correlation with other clinical pathologicalcharacteristics.Chi-square test showed that the expression of PDPN wasnot correlated with Bmi-1expression.chi-square test showed overexpression of Bmi-1correlated with higher T stages (p=0.017),and had acritical statistical significance to clinical stages (p=0.055), Bmi-1expression had no correlation with recurrence, lymph node metastasis,retropharyngeal lymph node metastasis;1patients occurred withnasopharyngeal recurrence,1case of patients with neck recurrence.15cases (15.5%) occurred with distant metastasis, mainly metastasis to bone,liver, lung, of which11cases (73%) for multiple organ metastasis;distant metastasis is the leading cause of death.The LRS、 DMFS、 DSS、OS、DFS for three years of the97patients were98%,85%,86%,85%,83%.Kaplan-Meier analysis revealed expression of PDPN has nothing to dowith the DMFS、OS、DSS、LRS of the patients;Kaplan-Meier analysisrevealed high expression of Bmi-1associated with decline DSS(p=0.041), the patients with high expression of Bmi-1has a higher chanceof metastasis, but there was no statistical difference (p=0.066); highclinical staging associated with DSS decrease (p=.045). Cox multiplefactor analysis indicated that the high expression of Bmi-1(p=0.036)associated with DSS, high expression of Bmi-1(p=0.041) associated withthe decline of OS;the clinical stage (p=0.047) associated with DSS;highexpression of Bmi-1(p=0.057), pharyngeal lymph node positive (p=0.059)had a critical statistical significance to DMFS decline.Conclusion:1. The expression rate of PDPN in nasopharyngealcarcinoma cells is low, which maybe reflect the pathological features ofnasopharyngeal carcinoma,PDPN may be used as a molecular marker todistinguish nasopharyngeal carcinoma with other head and necksquamous cell carcinoma.2. The expression rate of Bmi-1in nasopharyngeal carcinoma cell ishigh, Bmi-1can be used as a molecular marker of progression andprognosis of nasopharyngeal carcinoma.
