Objective:To summarize the clinical data of KD,and to investigatethe association between susceptibility to Kawasaki disease (KD) andpolymorphisms of both the interleukin17A gene (IL-17A) and the signaltransducer and activator of transcription gene (STAT3) in Han children inChongqing China.Methods: The clinical and laboratory data of KD in Han children inChongqing from2011to2012were retrospectively analyzed. The singlenucleotide polymorphisms of IL-17A (-197G/A,-692C/T) and STAT3(rs2293152C/G, rs957970C/T) in130patients with KD and147healthycontrol subjects were detected by sequence specific primers-polymerasechain reaction (SSP-PCR), and their differences were analyzed.Results:1、Occurrence rate of KD-CAL+was40.8%; The PLT and ESR inKD-CAL+were markedly increased than their in KD-CAL-〔PLT(×109/L)420.9±89.2vs355.2±68.2,ESR(mm/h)55.3±19.2vs39.8±10.2,P<0.05〕, and there were no significant difference between the two groups for WBC, CRP, Hb, Alb and LDH(P>0.05); Occurrence rate ofIVIG-resistant KD was8.5%; The Hb and Alb in the IVIG-resistant KDpatients were decline sharply compared with which in the IVIG-sensitive〔Hb(g/L)109.4±11.3vs91.6±8.5,Alb(g/L)36.7±3.6vs23.5±2.3,P<0.05〕, and there were no statistic differences between them forWBC, CRP, PLT, ESR and LDH(P>0.05).2、The polymorphisms of-197G/A in IL-17A:The frequencies ofAA genotype at-197G/A in IL-17A in KD patients (29.2%) wassignificantly higher than that in controls (15.0%)(P=0.004), and childrenwith A allele were1.412times more likely to suffer KD than children with Gallele (95%confidence interval1.009-1.975, P=0.044).3、The polymorphisms of-692C/T in IL-17A:There were no statisticdiffere nces in frequencies of genotype (CC、 CT、 T T) and allele (C、T) at-626in IL-17A between patients and controls (P>0.05).4、 The polymorphisms of rs2293152C/G in STAT3:The frequenciesof C allele and its homozygous CC genotype at rs2293152C/G in STAT3inKD patients were significantly higher than that in controls (P=0.006andP=0.002, respectively).5、 The polymorphisms of rs957970C/T in STAT3: There were nosignificant differences in frequencies of genotype (CC、 CT、 TT) and allele(C、T) at rs957970in STAT3between patients and controls (P>0.05). 6、There were no markedly differences in frequencies of genotype andallele between the KD-CAL+and KD-CAL-in IL-17A(-197G/A、-692C/T)and STAT3(rs2293152C/G、rs957970C/T); There were nostatistic differences could be found in data of laboratory test (WBC、Hb、PLT、CRP、ESR、Alb、LDH) between the genotype(P>0.05).Conclusions:1、The prevalence of KD-CAL+was40.8%and the IVIG-resistant KDwas8.5%in this study. The higher PLT and ESR were risk factors forKD-CAL+. The dramatic decline of Hb or Alb may be an alarm of IVIG-resistant in KD patients.2、The results indicated that the IL-17A gene promoterpolymorphisms at position-197G/A confered susceptibility to KD in Hanchildren in Chongqing China. The children with A allele were morevulnerable to KD. There were no significant correlation betweenpolymorphisms of I-692C/T in L-17A gene and susceptibility to KD.3、This study suggested that the association between STAT3genepolymorphisms at position rs2293152C/G and susceptibility to KD in Hanchildren in Chongqing China, and with C allele were more susceptibility toKD. G allele may play a role in preventing KD. But STAT3genepolymorphisms at position rs957970were possibly not related with KD inHan children in Chongqing China.
