| Objective:To compare the difference of serum sclerostin amongpostmenopausal women and the change of serum SO treated rhPTH(1-34),and investigate the relationship of serum SO and E2,bone mineraldensity,further to explorer the relationship of serum SO and Osteoporosis.Methods:95postmenopausal women were divided into normalBMD group(n=41,T>-1SD)and osteoporosis group(n=54,T≤-2.5SD,or fracture of one or more and T<-1SD)on the basis of whose bonemineral density.All participants were measured height,weight,body massindex(BMI),alkaline phosphatase(ALP),serum E2,serum calcium,serum phosphate and sclerostin(SO)by ELASA after collected fastingvenous blood and centrifuged.The patients in osteoporosis group weretreated with recombinant human parat-hyroid hormone (1-34),all thetreatments lasts twelve months.Serum sclerostin and other biochemicalindicators were measured in osteoporosis group after6months and12months treatment. Results:(1)The osteoporosis group in BMD,ALP,E2and So isdifferent with the normal BMD group:The level of the bone mineraldensity of lumbar vertebrae2-4,the bone mineral density of left femoralneck and E2were decreased by17.71%,26.37%and31.79%withcompared normal BMD group, Serum SO and ALP levels of theosteoporosis group were higher than those in the normal BMD group, andthe differences were significant(P<0.05~P<0.01),but there were nosignificant difference between the two groups of height,weight,BMI,serum calcium and phosphorus.(2)The serum E2and BMD of lumbarvertebrae L2-4and left femoral neck were negative correlation with SO[thecorrelation coefficients were r=-0.518(P<0.001),r=-0.225(P=0.045)andr=-0.221(P=0.49)]; Age and postmenopausal time were positivecorrelation with SO[the correlation coefficients were r=0.470(P<0.001)andr=0.406(P=0.001)].(3)The BMD of lumbar vertebrae2-4and left femoralneck and serum ALP were increased gradually with treated rhPTH after6months and12months with the rhPTH(1-34) treatment,and thedifference was especially(P<0.05~P<0.01).With the prolongation of thetreatment time increased,the serum SO was reduced gradually and thedifference was statistically significant(P<0.05~0.01);serum calcium,phosphorus with the prolongation of treatment time increased,but therewas no significant difference before and after the treatment,and E2had noobvious change. Conclusion:Serum SO was increased in postmenopausal women,it was related to E2and BMD,and was reduced gradually with treatedrhPTH. SO may participate in the development of postmenopausalosteoporosis. |
