| Objective: To study the role of JAK2/STAT3signalling pathwayplayed in invation and vascular mimicry of liver cancer cell QGY-7701invitro.Methods:After liver cancer cell were treated with5μmol/L and10μmol/L AG490(the inhibitor of JAK2)for48h, invating and forming tubesabilities of tumor cells are tested in vitro. with transwell and vascularmimicry experiment.RT-PCR was used to measure expression of Twist1andMMP-2mRNAs;Western blot was used to study expression ofp-STAT3,STAT3,Twist1and MMP-2proteins.Results: Compared to control group, less tumor cells permiatingmembrane and less tubes formed by liver cancer cells; RT-PCR show thatthe expression level of Twist1and MMP-2mRNA are decreased; Westernblot show that the expression level of p-STAT3,Twist1,MMP-2proteins aredecreased(P<0.05),while there are no difference of STAT3protein amongeach groups(P>0.05).Conclusion: The abilities of invation and vascular mimicry of livercancer cells can be inhibited by AG490in vitro, JAK2/STAT3signaling pathway promotes the abilities of invation and Vascular Mimicry of livercancer cells. |
