Objective:To investigate the potential protective effects of Shenmai(SM) combined with Ligustrazine injection on septic mice caused by cecal ligation and puncture (CLP), the relationship between two drugs and the mechanism of action that may be involved in.Methods:The model of sepsis was copied by the cecal ligation and puncture(CLP). Male mice were randomly divided into sham operation group,model group,Shenmai injection group,Ligustrazine injection group and Shenmai combined Ligustrazine injection group:Animals received a separate intraperitoneal injection of Shenmai (500mg/kg) and Ligustrazine injection (55mg/kg),or normal saline at24h,12h and1h before CLP.At24h after the model was copied, the ratio of wet weight to dry weight of lung tissue was tested,the changes of lung and kidney tissue were evaluated by HE staining. The activity of MPO,iNOS and the content of MDA,NO, BUN,Cr was detected by biochemistry enzymology.The changes of IL-6and TNF-a expression in mice serum was detected by ELISA. The expression of NF-κB and A20were determined by Western-blotting. Survival curves were obtained by the Kaplan-Meier method at72h after the model was copied.Results:(1) Compared with the sham-operative group, at each time point, the survival rate of mice in model group were lower;and the renal injury and lung injury were more serious;the levels of TNF-α and IL-6were higher.(2) Compared with the model group, in groups treated with Ligustrazine injection, Shenmai injection or Shenmai injection combined with Ligustrazine injection the survival rates of mice were improved; the renal injury and lung injury were reduced; the levels of TNF-a, IL-6,NO and iNOS, and the expression of NF-κB and A20were decreased; and the ratio of the expression of NF-κB and A20was recovered to close to the levels in sham group, especially in Shenmai combined Ligustrazine injection group.Conclusions:It is suggested that the sepsis could be caused by the cecal ligation and puncture, and Shenmai combined Ligustrazine injection have protective effects on sepsis-mice. Inhibiting the over expression of iNOS, preventing NF-Kb activating and decreasing systemic inflammation may be involved in the protective mechanism.
