PART1Analysis of siderophores genes in clinical isolates ofKlebsiella pneumoniaeObjective: To study the siderophores genes of clinical Klebsiellapneumoniae strains isolated from Children′s Hospital of Chongqing MedicalUniversity.Methods: A total of34clinical strains(13blood,21sputum)ofKlebsiella pneumonia isolated from hospitalized patients were selectedrandomly. PCR was used to detect the siderophores genes.Results: Of the34strains, all strains carry enterobactin (Ent) gene(entB),9strains (26.5%) with yersiniabactin (Ybt) gene (fyuA),3strains(8.8%) with salmochelin, glycosylated-Ent (Gly-Ent) gene (iroN), and1strain (2.9%) with aerobactin (Aer) gene(iutA).25strains (73.5%) containEnt gene only, and9strains (26.5%) also contain other siderophores genesbesides Ent. Both entB and ybtS were positive in6strains (17.6%),all of theentB, ybtS and iroN were positive in2strains (5.9%), and only1strain (2.9%) had the4siderophores genes.Conclusion: The siderophores gene of the clinical Klebsiellapneumoniae strains is Ent gene primarily in our hospital, and the detectionof nonenterobactin siderophores genes should be noticed. PART2The reactive oxygen species up-regulation theexpression of NGAL in A549cell which infected byKlebsiella pneumoniaobjective: To study the effect of reactive oxygen species (ROS) to theexpression of neutrophil gelatinase-associated lipocalin(NGAL), an naturalimmune protein, in A549cell lines which infected by Klebsiella pneumonia(KP).Methods: The A549cell lines were divided into5groups forexperiment: normal control group, H2O2group, H2O2+vitamin E (VE)group, KP group and KP+VE group. The level of ROS were measured byluminol chemiluminescence, NGAL mRNA expression were detected byquantitative real-time PCR (qRT-PCR), NGAL protein expression weredetected by Western blotting.Results: The level of ROS of the KP group and H2O2group were higherthan the normal control group, meanwhile, the NGAL mRNA and proteinexpression of the two groups in A549cell were higher than the control group.But when the VE was added, the level of ROS of the two groups weredecreased, accordingly, the NGAL mRNA and protein expression of the twogroups were also declined.Conclusion: The reactive oxygen species up-regulation the expressionof NGAL in A549cell lines, when the A549cell lines were infected by Klebsiella pneumonia.
