Objective:To explore how inflammatory factors and apoptotic cellsregulate antigen-presenting cells (APCs) related cytokine—IL27expression by phagocytosing apoptotic cells.Methods:Measure and compare the luciferase activity of RAW264.7cell transfected with IL-27p28promoter, treated with different stimulus.Using QRT-PCR measure and compare IL-27p28subunit mRNArelatively expression in RAW cells, mice peritoneal macrophages (PMs),microglia cells and bone marrow derived dendritic cells (BMDCs), treatedwith LPS, INF-γ and X-ray induced apoptotic cells. Western blot wasperformed to detect IL-27expression in different stimulus groups. ELISAswas performed to detect IL-27expression in different stimulus groups inmicroglia cells’ medium.Results:The expression of IL-27p28subunit mRNA and IL-27proteinwas up-regulated when APCs were treated with LPS, the expression ofIL-27p28and IL-27had dose dependent effect with LPS. APCs IL-27p28subunit mRNA and IL-27expression was significantly up-regulated whencells were treated with IFN-γ plus LPS (P<0.05). The expression of IL-27 p28mRNA and IL-27was significantly down-regulated when APCs waspre-treated with apoptotic cells (P<0.05).Conclusion:LPS and IFN-γ can up-regulate IL-27expression in APCs,IL-27expression was suppressed when APCs phagocytosis of apoptoticcells.
