Research Of Carboplatin-PECE Hydrogel Pharmacokinetic In Rat Abdominal Cavity

Objective: Injected PECE hydrogel drug slow-release system into rat abdominal cavity，through a reverse HPLC method to determine the different time’s concentrations ofcarboplatin periods in the rat plasma, to analyze its pharmacokinetic characteristics,provide theoretical basis for Carboplatin-PECE hydrogel slow-release systemintraperitoneal chemotherapy.Methods: Preparation of carboplatin-PECE hydrogel;18healthy female Wistar rats wererandomly divided into normal saline group and carboplatin group and carboplatin-PECEhydrogel group. Through a reverse HPLC method to determine carboplatin concentrationafter the treatment in plasma of rats each10min,30min,1h,2h,3h and4h, to analyzeits pharmacokinetic characteristics and compare the side effects of drugs.Results: Carboplatin-PECE hydrogel at low temperature can be injected sol state, in37℃is not flowing gel state, after the carboplatin solution injected into the rats abdominalcavity,determination of10min,30min,1h,2h,3h and4h carboplatin group of plasmadrug concentration, respectively is:0.883±0.0036mg/ml、0.982±0.0051mg/ml、0.499±0.0046mg/ml、0.125±0.0002mg/ml、0.031±0.0006mg/ml、0.007±0.000005mg/ml,Carboplatin-PECE hydrogel group drug concentration at the same time, respectively is:0.720±0.0003mg/ml、0.810±0.0023mg/ml、0.615±0.0034mg/ml、0.151±0.0023mg/ml、0.037±0.0002mg/ml、0.0090±0000006mg/ml. According to compare two groups of datadifference have statistical significance (P <0.05). The under area of Carboplatin group’stime curve is about1.0940±0.002mm, the under area of Carboplatin-PECE hydrogelgroup’s time curve is about1.1116±0.003.According to compare two groups of datadifference have statistical significance (P <0.05). For important organs of rats, PECEhydrogel did not increase the side effects of carboplatin.Results: As a kind of injection drug slow release system, PECE hydrogels have goodsustained release characteristics. Can be loaded by anticancer drugs-Carboplatin slowly released into the rats abdominal cavity and then absorbed into the bloodstream andmaintain a relatively high concentrations, for Carboplatin-PECE hydrogel intraperitonealchemotherapy to lay the theoretical foundation.
