Abstract/Summary:
With the widely application of nanomaterials, the potential biological security of thenanomaterials has caused the scientific research workers’ attention. Hydroxyapatite (HA) asgood biomaterials which were widely used in bone repair and drug delivery, so it is worth topay close attention to the biological security of these materials. In this study, we were usedthe defect-related luminescence HA nanoparticles as a study subject, based on thecharacterized of the SEM, XRD, DLS methods, and use the MC3T3-E1cells as a cellmodels to study the biological effect. Though the cell viability test, the uptake and locateinto cells, the LDH release test, the intracellular reactive oxygen species test andmitochondrial membrane potential test method to study the biological effect of thisnanoparticles. Besides that, we have performed the DNA gel electrophoresis and Caspase-3gene level test to recover the mechanism of the potential cytotoxicity on the molecules level.The results indicating that nano HA can uptake into by pinocytosis way and mainlylocalization in the lysosome; the intracellular nanoparticles and induce the ROS level overexpress and decrease the mitochondrial membrane potential, so that to inhibit the cellproliferation. The further study indicating that the nano HA can induce the damage of DNAand arrest the cell cycles at S phase, through the activity of caspase-3and induce cellsapoptosis.
