Study On Polymorphism Of IL-18of Qinghai Tibetan People’s HBV Susceptibility Genes

【Objective】To the detect IL-18’s gene polymorphism of Tibetan HBV patient inQinghai.,To discuss the reason of the high rate of infecting HBV in Tibetan andIL-18playing the role in susceptibility of HBV and provide the basis for the diagnosisand treatment.【Methods】Choose the local Tibetan residents and extract10ml blood of230hepatitis B patients.According to the result of testing five items of hepatitis B,choosethe134cases of healthy people to compare with HBV patients in the same age andgender.In view of polymorphism site of IL-18,screening the distribution frequency ofTibetan people with the way of PCR-SSCP and appraise it with sequencinggene.Using the statistics method to analyse IL-18of screening, and judge whetherthere is obvious difference or not,with way of counting the distribution frequency ofIL-18between Tibetan HBV patient and healthy people.Compare and analyse SNPand the relationship of each type of blood serum.To analyse the obtained data withstatistical software of SPSS17.0, such as genotype frequency and frequencycomparison of allele、estimation of relative risk.【Result】1.137G/C:the study group and control group(1)The compare of three genes’frequency: X~2value is1.079,P>0.05. The compare of allele’s frequency: X~2value is0.123,P>0.05.(2)The analysis of logistic:OR value of GG gene is0.953,95%CIinterval is0.517~1.757;:OR value of CG gene is0.980,95%CI interval is0.529~1.816,OR value of allele is1.110,95%CI interval is0.619~1.990.2.-607C/A: the study group and control group(1)The compare of three genes’frequency: X~2value is1.150.,P>0.05.The compare of allele’s frequency: X~2value is1.139,P>0.05.(2)The analysis of logistic:OR value of CC gene is1.169,95%CIinterval is0.747~1.830.:OR value of CA gene is1.046,95%CI interval is 0.685~1.598；OR value of AA gene is0.772,95%CI interval is0.471~1.267. ORvalue of allele A is1.176,95%CI interval is0.873~1.585.3.-105A/C: the study group and control group(1)The compare of three genes’frequency: X~2value is1.615,P>0.05. The compare of allele’s frequency: X~2value is1.615,P>0.05..(2)The analysis of logistic:OR value of AA gene is0.925,95%CIinterval is0.510~1.679; OR value of AC gene is.0981,95%CI interval is0.537~1.793; OR value of allele C is1.166,95%CI interval is0.663~2.05.【Conclusion】There is no obvious correlation betweem polymorphism of IL-18’sSNP s’-607C/A、-137G/C、105A/C and susceptibility of hepatitis b.
