ObjectiveResearch the separation and purification of bone marrow mesenchymal stem cells in vitroculture by tissue engineering, and guide into cartilage cells for injecting into articular cavity, treat thearticular cartilage defect caused by adjuvant arthritis in rabbit. Observe the effect of the treatment,and then discuss the results of cartilage repair by autologous chondrocytes in vitro cultured. Toprovide corresponding theoretical basis and experimental basis for clinical use of tissue engineeringtreatment rheumatoid arthritis.Methods⑴Divided the rabbits into three groups: control group (8), arthritis model group (8), cartilagecells-scaffold complex treatment group(8).⑵Inject adjuvant into the rabbits’ knee of model group and treatment group to induced arthritis,and physiological saline into control group.⑶Extract the bone marrow of treatment group, and then separate the bone marrowmesenchymal stem cells. Cultivate and purification of it in vitro, induce it to cartilage cell.⑷Mix the cartilage cells and scaffolds. Observe the growth of cartilage cells within the stentby electron microscopy, and inject it into articular cavity. Wait for a month. Inject physiologicalsaline into control group and model group.⑸Obtain three groups of the rabbits’ knee joints for gross observation. Detect the curativeeffect of cartilage cells–scaffolds. Do some histologic grading and HE staining andimmunohistochemical examination.Results⑴All of the rabbits injected by adjuvant have deformity of joints, to prove the successful ofmodels’ foundation. ⑵The bone marrow mesenchymal stem cells in vitro change their form after the addition ofinductor. The results of Alcian blue and Type II collagen immunohistochemical can prove that thecells induced are cartilage cells.⑶The analysis of pathology shows that the situation of cartilage of treatment group is muchbetter than model group.ConclusionDue to the pathology analysis of articular cartilage defect in treatment group, we prove thattissue engineering is a effective method in treating adjuvant arthritis.
