ã€Objective】 Chang liver cell is a hepatoma cell line, whether it can be applied for BALshas not been reported. To search for a novel cell source, this study evaluated the hepatocyticfunction of Chang liver cells both in vitro and in vivo.ã€Methods】 To evaluate the hepatocytic function of Chang liver cells in vitro, theexpression of specific hepatocyte markers were detected by western-blotting and laserconfocal microscopy. Related gene expression of hepatic function was detected by RT-PCR.The acute liver failure (ALF) model was established by90%partial hepatectomy. Changliver cells were intra-splenically implanted24hours before the surgery and the serumsamples were collected per24hours to test the liver functions. And the survivals wereassessed.ã€Results】 In vitro, Western blotting and laser confocal microscopy showed conspicuousexpression of hepatocytic markers, such as albumin (ALB), uridine diphosphateglucuronosyltransferase (UGT) and cytochrome P4503A4(CYP3A4) in Chang liver cells.RT-PCR also exhibited the expression of related genes at mRNA level. In vivo, the survivalof rats receiving Chang liver cells was significantly improved and reached40%against0%in controls (P<0.01). Liver functions of rats receiving Chang liver cells were alsosignificantly improved24h after ALF, which was evidenced by the decreased levels ofalanine transaminase (ALT), aspartate aminotransferase (AST), bilirubin and alkalinephosphates (ALP) in blood samples of rats implanted with Chang liver cells (ALT,619.33±85.64vs.1798±424.06μ/l, G2vs.G1; AST,1732.83±162.37vs.3033.25±506.73μ/l,G2vs. G1;ALP,330.33±26.18vs.511.6±40.25μ/l, G2vs.G1; p<0.05).ã€Conclusion】Chang Liver cells, expressing hepatocytic markers, significantly improvingliver function and survival after ALF, can serve cells for liver support systems. |
