| Objective:Renal cell carcinoma (RCC) is a common cancer of urinary system, but the molecular mechanisms that underlie RCC are not fully understood. Fibulin-1, a multi-functional extracellular matrix protein, has been demonstrated to be involved in many kinds of cancer, while its function in RCC is unclear. So here we investigated the expression and function of Fibulin-1in RCC.Methods:Real-time PCR, Western blot analysis and immunohistochemistry were applied to determine the expression of Fibulin-1in RCC cells and patient tissues respectively. When overexpressing fibulin-1in RCC cells, we investigated its impact in some aspects such as cell proliferation,apoptosis,movement and tumorigenicity.Result:Fibulin-1was significantly down-regulated in RCC cell lines and patient tissues, and its dysregulation was associated with RCC progression. Overexpression of Fibulin-1dramatically led to reduced cell growth, enhanced tumor cell apoptosis, and decreased cell motility in cultured RCC cells and xenograft tumor in nude mice.Conclusion:Fibulin-1is down-regulated in RCC and shows a significant function of tumor suppressor in RCC. |
