Objective:To study the effect of imipenem/cilastatin sodium on quorum sensing defective Pseudomonas aeruginosa (C84) and Pseudomonas aeruginosa PAO1inrespect of Alginate and Alginate Biosynthesis Gene Expression.Methods:Tube fold dilution was used to detect the minimum inhibitory concentration (MIC) of imipenem/cilastatin on PAO1and C84. Modified plate culture method was used to establish the biofilm model in vitro. The effect of1MIC and10M IC imipenem/cilastatin on PAO1and C84Alginate Biosynthesis was observed by try ptophan reaction and semi quantitative RT-PCR was used to determine the effect on the gene expression of algD.Results:When PAO1and C84biofilm were exposed to1MIC imipenem, algD expression level were higer in the test group in the three time point compared to the controls;When exposed to10MIC imipenem,both PAO1and C84had less algD expression at any of the three time points than the controls.When exposed to1MIC and10MIC imipenem,the production of alginate had a peak at the3rd day in both PAO1and C84.conclusion:Imipenem/cilastatin can induce algD expression of the wild-type and quorum sensing system deficient Pseudomonas aeruginosa, QS-deficient P. aeruginosa have less algD levels than the wild-type. algD expression and alginate synthesis reached a peak on the third day.
