ObjectiveSo far there are only a few reports about the express of RhoGDI2and PAK2(itssuspected downstream target protein) and their Correlation with the clinicopathologicalcharacteristics of a large sample in human gastric cancer. In this study, we intend to use theimmunohistochemical techniques to detect the expression of RhoGDI2and PAK2in103cases of gastric cancer. And we intend to detect the correlation with the clinicopathologicalcharacteristics. Assess the clinical significance of RhoGDI2and PAK2in the invasionand metastasis of gastric cancer.MethodsWe use the American Joint Committee of Cancer (AJCC) TNM staging standard ofgastric cancer (2010, seventh edition). The expression of RhoGDI2and PAK2in103casesof Gastric cancer tissue were investigated by immunohistochemical EliviSionTMplustechniques. All statistical data was Performed by SPSS18.0software. P values less than0.05Were considered statistically significant.Results1. RhoGDI2was mainly expressed in the cytoplasm or the nucleus.The expressionrate of RhoGDI2protein in gastric cancer was72.82%(75/103), well as the rate ofnormal tissues was0.00%(0/21),the difference was statistically significant (χ~2=38.70,p<0.05), The positive rate of poorly differentiated gastric cancer group was significantly higher than the high-middle differentiated gastric cancer group (χ~2=6.37, p<0.05);Theinvasion grade of gastric cancer was significant difference (χ~2=21.64, p<0.05); The groupof lymph node metastasis was significantly higher than those without metastasis (χ~2=54.50,p <0.05); the group of distant metastasis was significantly higher than those withoutmetastasis (χ~2=19.81, p<0.05);the group of TNM stage Ⅲ+Ⅳ was significantly higherthan Ⅰ+Ⅱ(χ~2=24.90, p<0.05),The expression rate of RhoGDI2was not statisticallysignificant difference (p>0.05) in gender, age(<60years or≥60years),expression of tumorlocation (gastric cardia and gastric fundus, gastric body, gastric angle, antrum),size of theprimary tumor (<5cm or≥5cm).2. PAK2was mainly expressed in the cytoplasm. The expression rate of PAK2protein in gastric cancer was60.20%(62/103), well as the rate of normal tissues was0.00%(0/21),the difference was statistically significant (χ~2=25.28, p<0.05), The positive rate ofpoorly differentiated gastric cancer group was significantly higher than the high-middledifferentiated gastric cancer group (χ~2=7.98, p<0.05);The invasion grade of gastric cancerwas significant difference (χ~2=7.98, p<0.05); The group of lymph node metastasis wassignificantly higher than those without metastasis (χ~2=22.02, p<0.05); the group of distantmetastasis was significantly higher than those without metastasis (χ~2=4.01, p<0.05).;thegroup of TNM stage Ⅲ+Ⅳ was significantly higher than Ⅰ+Ⅱ(χ~2=17.30, p<0.05),Theexpression rate of PAK2was not statistically significant difference (p>0.05) in gender,age(<60years or≥60years),expression of tumor location (gastric cardia and gastric fundus,gastric body, gastric angle, antrum),size of the primary tumor (<5cm or≥5cm).3. The relationship of the in situ expressions of RhoGDI2and PAK2were positivecorrelation (χ~2=5.33, p<0.05). Conclusionthe expression of RhoGDI2and PAK2are closely related with the invasion andmetastasis and pathological features of gastric cancer. The activation of PAK2is closelyrelated with RhoGDI2. PAK2is most likely one of the downstream target proteins ofRhoGDI2. RhoGDI2and PAK2may be involved in regulating the process of invasion andmetastasis of gastric cancer.
