Objective: To explore normal tissues and tumors of the gastrointestinal tract in MLH1`gene exon2methylation status, and analyze its relationship with MLH1promotermethylation.Methods:The bisulfite genomic sequencing was used to analyse the CpG sites of the5end (promoter, exon1and intron1) of the MLH1gene in colorectal cancer tissues,and gastric cancer tissues.Results: In this experiment, normal human peripheral blood, normal tissue and acancer adjacent tissue as control samples, two cancer cell lines and five colon cancertissues and three gastric cancer tissues as experimental samples. Extraction DNA andDNA conversion for CT, after conversion of the DNA as a template for sequencingPCR after bisulfite modification (i.e., the BSP). Finally, the PCR products weremolecular cloning, sequencing of the cloned product discussion to analyze the results.In the adjacent normal tissue and cancer tissues, exon2shows a high level ofmethylation, but in the tumor tissue or cells outside exon2demethylation tendency,and the surrounding area demethylation phenomenon obviously.Conclusion: Demethylation of the region around exon2of MLH1gene may relatewith its promoter methylation in Gastrointestinal cancer.
