| Objective The aim of this research is to assessing the efficency of the bFGF-collagen integrated with PRP (platelet-rich plasma) in the long bone defects models.Method Firstly, platelet-rich plasma was extracted from the blood of the male rabbits;secondly, The80defects were randomized into4groups and the defects were treatedwith nothing (group A), simple collagen scaffolds (group B), collagen scaffoldsintegrated with PRP(group C) and bFGF-collagen scaffolds integrated with PRP (groupD). Thirdly, five rabbits of each group were sacrificed at4and8weeks after surgery(n=10) and the plain X-ray was performed. The specimens were fixed with10%paraformaldehyde for Histological observation. Bone formation was assessed in eachgroup under a protocol with triple blinding according to Lane–Sandhu X-ray scores inX-ray results. In addition, the micrographic images of histologic section were analysedwith Image-Pro Plus image analytical software and the areas of newly formed bone werecalculated per total bone defect area. Results The defects treated with bFGF-collagenscaffolds integrated with PRP (group D) showed significantly higher scores by X-rayanalysis in any time points. And more bone formation determined by both histology thanother three groups after4and8weeks (p<0.05). both B and C significantly higher thangroup A (p<0.05).Conclusions the scaffolds of bFGF-collagen integrated with PRPmay be an effective approach to promote repair of segmental bone defects and have greatpotential for repairing large segmental bone defects in clinic. |
