Objective To study whether the synthesized human cell penetrating peptide hCPP10can permeate the blood-brain barrier, their distribution differences in SD rat brain and theireffects on the cell viability.Methods We test whether His-hCPP10-GFP can penetrate the blood brain barrier ofSD rats under fluorescence microscope; we test the distribution of his-hCPP10-GFP inSD rat brain also under fluorescence microscope; we compare the effects on cell viabilityof His-hCPP10-GFP and His-TAT-GFP by MTT.Results (1) The acquired of the fusion protein His-hCPP10-GFP, His-TAT-GFP andHis-GFP;(2) the His-hCPP10-GFP was detected can effectively penetrates blood-brainbarrier in SD rats;(3) the His-hCPP10-GFP was detected mainly gathered in the cerebralcortex, basal ganglia, hippocampus and periventricular area in SD rats;(4) the effects onthe viability of cells of His-hCPP10-GFP is much lower than that of his-TAT-GFP.Conclusion Human cell penetrating peptide hCPP10can effective permeate the bloodbrain barrier, mainly gathered in the cerebral cortex, basal ganglia, hippocampus andperiventricular area and their effects on the viability of cells is much lower than that ofTAT.
