Objective:To observe the intervention effect of compatibility of SALVIANOLICACID B、C on proinflammatory cytokines CCL2、CCL3of renal fibrosis in rats.Methods:50SD rats were randomly divided into5groups: normal group,modelgroup, Salvianolic acid B group, Salvianolic acid C group, Salvianolic acid B+C group.In addition to the normal group, the rest all caused by unilateral ureteral obstruction(UUO) rats models, that ligature the right ureter after anesthesia. Normal group andmodel group were treated with the water. Other treated groups were given relevantdrugs for14days by gavage. The fasting blood of each rat was used to determine therenal function(Serum creatinine, Serum blood urea nitrogen) and metabolic cages wereused to collecting urine for β2-Microglobulin(β2-MG) and N-acetyl-β-D-glucosami-nidase (NAG). The nephridial tissues were made into paraffin section by dehydrationand embedding which were used for HE staining, Masson staining, and detected type Icollagen (ColⅠ)、type Ⅲ collagen (Col Ⅲ) and α-smooth muscle actin (α-SMA) byimmunohistochemistry staining. Inflammatory chemokine CCL2and CCL3proteinwere analyzed by Western Blot.Results:1. Tubulointerstitial pathological changes (1) HE staining There werenot any pathological changes in normal kidney of normal group.UUO groups showed alarge number of atrophic tubular epithelial cells (particularly the tubular in cortex andrecent marrow) with vacuolar degeneration, luminal expansion, and inflammatory cellsinfiltration.The pathological changes in the treatment groups improved ameliorate invary degrees. The scores of HE in salvianolic acid B、B+C group were lower than insalvianolic acid C group(P<0.05).(2) Masson staining Deposition of collagen inrenal tissue was not obvious in normal group. Light blue collagen could be seen in renal capsule, base material of nephric tubules, and mesenchyme in UUO modelgroups. Collagen deposition in all the treatment groups is lighter than the model group.The scores of Masson in salvianolic acid B+C group were decline than in salvianolicacid C group (P<0.05).2. Determination of serum Scr and BUN The level of serum Scr and BUN in modelgroup was significantly higher than in the normal group (P<0.05). The level of serumScr in salvianolic acid C treatment group wassignificantly decline than the modelgroup (P<0.05). The level of serum Scr in salvianolic acid C treatment group wassignificantly decline than in salvianolic acid B+C group (P<0.05).3. Results of the β2-MG and NAG The level of β2-MG and NAG in model group wassignificantly higher than the normal group (P<0.05). The level of β2-MG、NAG intreatment group was significantly decline than the model group (P<0.05).4. Results of immunohistochemistry of cytokines in nephridial tissue The level ofColⅠ、Col Ⅲ、α-SMA in model group was significantly higher than the normal group(P<0.05). The level of ColⅠ、Col Ⅲ in salvianolic acid B、B+C group and α-SMA intreatment group was significantly decline than the model group (P<0.05). The level ofColⅠ in salvianolic acid B、B+C group was significantly decline than in salvianolicacid C group (P<0.05). The level of Col Ⅲ、α-SMA in salvianolic acid B+C group wassignificantly decline than in salvianolic acid C group (P<0.05).5. Results of CCL2and CCL3in kidney tissue by Western-Blotting The level ofCCL2and CCL3in model group was significantly higher than the normal group(P<0.05). The level of CCL2、CCL3in treatment group was significantly decline thanthe model group (P<0.05). The level of CCL2in salvianolic acid B+C group weresignificantly lower than in salvianolic acid B、C group (P<0.05).Conclusion: Compatibility of Salvianolic Acid B、C may protect renal function,improve tubular function and renal pathology in unilateral ureteral obstruction in acertain degree,deposit of Col I、Col Ⅲ and α-SMA would be alleviated. The mechanism may interfere with UUO rat kidney with the secretion of inflammatorychemokine CCL2and CCL3.
