Objective: To investigate the potential effects of tranilast (Tranilast, TRA) on co-relatedchanges of IL-4, IL-13expression and pulmonary fibrosis in rats with paraquat (Paraquat,PQ) intoxication for providing basis theory of effective treatment of paraquat poisoning.Methods:90male SD rats were randomly divided into normal control group (N group),model group (P group), dexamethasone treatment group (D group), tranilast treatment (Tgroup), dexamethasone combined with tranilast treated group (DT group) with18animalsin each group. PQ (30mg/kg) was administered to create PQ poisoning models, while thenormal control group were given normal saline of the same quantity. The poisoning ratswere treated with dexamethasone (1mg/kg/d) by intraperitoneal injection for7days, andthen adjusted the dosage of0.5mg/kg/d for further treatment. Tranilast (250mg/kg) weregiven by gavage once a day. All the rats were sacrificed to isolate lung tissues on the7th,14thand the21stday respectively and detected the pathological changes focused onpulmonary fibrosis by HE and Masson staining, then analyzed the level of IL-4and IL-13by ELISA in serum.Results: Compared with other groups, the level of IL-4and IL-13increased significantly,and the most significant degree of pulmonary fibrosis were observed in P group. Comparedwith other groups, the level of IL-4and IL-13decreased significantly, the degree ofpulmonary fibrosis ameliorates significantly. Comparisons between D group and DT group,the level of IL-4and IL-13and the degree of pulmonary fibrosis were changed in someextent in the P group. Comparative differences among groups were statistically significant(p <0.05).Conclusion: There are positive correlations between IL-4, IL-13levels and pulmonaryfibrosis. Therefore, tranilast may be viewed as an effective substitute to treat paraquatpoisoning, which play a vital role in alleviating pulmonary fibrosis.
