Objective: To investigate the expressions of TGF-β1and α-SMA in the model of biliarytract injury in rabbits intervened by the PNS, in order to judge whether the PNS couldinhibit the proliferation of fibroblast, thus inhibiting bile duct injury after repair of scarcontracture and stricture.Methods:75domestic rabbits were randomly divided into a negative control group, thepositive control group and experimental group in which were established into the model ofbiliary tract injury. The negative control group was extracted bile duct tissues respectivelyafter operation of the model1week,3weeks and1month.Positive control group wasinjected with the6mg/Kg/d of salvia miltiorrhiza for one week, and then extracted bileduct tissues with1week,3weeks and1month. Experimental group was divided into A, Band C three subgroups after operation,each subgroup was intervened for one weekrespectively with the4mg/Kg/d,8mg/Kg/d and16mg/Kg/d of the PNS injection, and thenextracted bile duct tissues with1week,3weeks and1month after administration. HEstaining was used to observe the morphological comparison of each group under lightmicroscopy. Immunohistochemical staining was performed to detect the the expressions ofTGF-β1and α-SMA in biliary tract scar tissues which was intervened with different drugconcentrations and periods. the data were analyzed statistically using the SPSS19.0software.Results: Immunohistochemistry shows:the expressions of TGF-β1and α-SMA in thenegative control group gradually increase along with the prolong of biliary tract injurytime(P <0.05), and the same as in the positive control group which was intervened bysalvia miltiorrhiza injection(P <0.05). The expressions of TGF-β1and α-SMA graduallydecreased at the same period with the increasing of the PNS injection concentration in comparison between the subgroup of A, B and C(P <0.05), while the expressions ofTGF-β1and α-SMA gradually increased within the subgroup to compare the sameconcentration of PNS injection as the prolong of biliary tract injury time(P <0.05).compared with the negative control group, The expression of TGF-β1and α-SMAdecreased significantly in group A, B, C(P <0.05).compared with the positive controlgroup, The expression of TGF-β1and α-SMA decreased significantly in group C (P <0.05).Conclusion:1.PNS maybe inhibit the proliferation of fibroblast.2.PNS on fibroblastinhibitory effect is positively correlated on the dose-dependent manner, and negativelycorrelated with time relationship.3.The PNS injection on biliary tract scar inhibitory effectis better than that of salvia miltiorrhiza injection.
