Objective:To investigate the roles of Hedgehog signaling pathway in prolifera-tion andimmigration of HepG2.2.15cell.Methods: HepG2,HepG2.2.15cells were divide into blank group,and the low, median andHigh concentration of cyclopamine treatment groups,which was treated with theConcentrations of cyclopamine were5μmol/L,15μmol/L,25μmol/L,respectively. After24h,48h,72h, The proliferation of HepG2.2.15cell was detected by MTT.The cellular DNAsynthesis were detected by EdU. The invasive ability was measured by Boyden chamberinvasive models.The migration ability was measured by scratch test. The expression levelsof Gli1,MMP-9mRNA were measured by rea1-time PCR.Results:1. MTT assay results showed that the cyclopamine treatment groups with theconcentration of5μmol/L、15μmol/L、25μmol/L was significantly reduced cellproliferation compared with the control group,which the difference was statisticallysignificant (p<0.05).2.EdU detection results showed the cyclopamine treatment groupsdecreased the cellular DNA synthesis compared with the control group,Individually.whichthe difference was statistically significant (p<0.05).3.The ability of metastasis andinvasion by transwell and scratch test, which differences have statistical significancecompared with the control group(p<0.05).4.Real-time PCR results showed Gli1of thecyclopamine treatment group with the Concentration of5μmol/L、15μmol/L、25μmol/Lwere cut down by50.4%,61.2%and75.7%on mRNA level, Respectively.5.Real-timePCR analysis of MMP-9showed that its expression was decreased on mRNA levels whichthe differences had statistical significance compared with the control group (p<0.05).Conclusions: The effect of cyclopamine can reduce the migration and proliferation inHepG2.2.15cells.And show asignificantly Negative correlation.Hedgehog signal-ingpathway may play an important role in the migration and proliferation of hepatoc-ellularcarcinoma by up regulating MMP-9mRNA expression....
