Objective: Breast cancer is the most common cancer among women and leading cause ofdeath worldwide, and its occurrence, development is a more steps and multiple genesregulated processes, In breast cancer, expression of PDGF in tumor cells and PDGF receptorsin stromal cells have been reported. PDGF receptors are also expressed in the tumor cells,which correlate with tumor progression and invasion. So it is very important to investigatedthe molecular mechanisms of PDGF on affecting biological behavior in breast cancer cells byusing MTS assay, cell count experiment and the cell migration assay, which have bearing onthe occurrence, development, progression and invasion of mammary carcinoma.Methods: The viability of SK-BR-3mammary carcinoma cells by cultured were treated withdifferent concentrations of PDGF-BB(0.5、1、2、4ng/mL) in logarithm growth stages wasdetermined by CellTiter96AQueousOne Solution Cell Proliferation Assay Kit (MTSassay) based on the optical density values measured at490nm, migration distance of eachmass. concentration were measured0h,12h,24h,48h,calculate the average migration distancefor each mass. concentration,cell count experiment and Cell Wound Healing Assay by cellmigration assay. All date were analyzed using SPSS18.0, One-way ANOVA and Two-wayANOVA was carried out on the experimental data. P<0.05was considered statisticallysignificance.Result: Compare with SK-BR-3mammary carcinoma cells without any drug, The PDGF-BBof4ng/mL promotes obviously the migration and proliferation of SK-BR-3mammary cells.The promoting was positively associated with drug concentration. With the increase ofstimulation time and concentration, PDGF-BB on SK-BR-3breast cancer cells promotemigration and proliferation can be strengthened in time-dependent and concentration-dependent manner.Conclusions: The PDGF-BB can promote migration and proliferation of the human SK-BR-3Mammary Carcinoma Cells obviously in a specific concentration range. This provideevidence for a novel role of PDGF-BB in tumor growth and spread of human breast cancer. Itcan be used as a prognostic marker of breast cancer in clinical practice.
