Objective:To investigate the neuroprotective effects of Gentiopicrside、Phillyrin and Mollugin on1-Methyl-4-phenylpyridinium (MPP+)-induced injury in SH-SYSY neuroblastoma cells line.Methods:SH-SY5Y cells were incubated with Gentiopicrside、Phillyrin and Mollugin in different concentrations. MTT metabolic rate was measured as the cell survival rate in order to determine the effective concentration. After SH-SY5Y cells were pre-incubated with Gentiopicrside or Phillyrin (100,10and1μmol·L-1) for24hours prior to exposure to MPP+(1mmol·L-1) for48hours, the MTT metabolic rate and LDH leakage rate was observed.Results:Gentiopicrside or Phillyrin could not decrease the survival rate of SH-SY5Y cells at the concentration from1to100μmol·L-1, thus, we could determine the selected drugs concerntration. Mollugin significantly inhibited cell activity of SH-SY5Y cells. Compared with the control group, MPP+decreased the MTT metabolic rate (p<0.001). However, Gentiopicrside significantly increased the cell activity (p<0.01) and decreased LDH leakage rate (p<0.05) of SH-SY5Y cells after MPP+induced injury; Phillyrin significantly increased the cell activity (p<0.05) and decreased LDH leakage rate of SH-SY5Y cells after MPP+induced injury.Conclusion:Gentiopicrside or Phillyrin protects SH-SY5Y cells against MPP+-induced cytotoxicity, anti-inflammatory, reducing oxidative stress and the subsequent anti-apoptotic effect may be the underlying mechanisms for the neuroprotective effects of Gentiopicrside or Phillyrin. To provide the basis for an in-depth to explore Gentiopicroside or Phillyrin neuroprotective effects of Parkinson’s disease.
