PurposesTo investigate the expression characteristics of Src and VEGF in the human primaryCNS Diffuse large B cell lymphoma (PCNSL-DLBCL) and their clinical pathologicalsignificances.Method and Materials1. The specimen obtained from the archives of Institute of Pathology, Tongji Hospital,Tongji Medical College, Huazhong University of Science and Technology withdiagnosis of primary CNS DLBCL.10cases of non-CNS DLBCL were used as thecontrol group.2. Immunohistochemistry was used to detect the expression of Src and VEGF in thePCNS-DLBCL.3. The positive expression of Src and VEGF in the lymphoma cells of PCNS-DLBCLwas semi-quantified as positive expression index (PEI) and recorded. Thedifferences between the PCNS-DLBCL and non-CNS-DLBCL groups wereanalyzed and also combined with the previous data of Microvascular Density(MVD) in our laboratory. The t-test was used to detect the differences betweenthe two groups, and the simple correlation analysis was performed within each group.ResultsThe Src and VEGF both were positively expressed in the cytoplasms andcytoplasmic membranes of lymphoma cells of PCNS-DLBCL and non-CNS-DLBCL.There was no significant difference in PEI of Src between these two groups (P>0.05).However, the PEI of VEGF in PCNS-DLBCL group was significantly higher,compared with that in the non-CNS-DLBCL group (P<0.05).In PCNS-DLBCL group, the analysis showed that the Src PEI was correlated withthe changes of MVD detected by CD34immuno-labelling (P<0.05), but not with thatof MVD by CD31immuno-labelling (P>0.05). The VEGF PEI was not correlated with the changes of Src PEI, CD34labelled MVD and CD31labelled MVD. Innon-CNS-DLBCL group, no significant correlation were found between the Src PEI,VEGF PEI, CD34labelled MVD and CD31labelled MVD.ConclusionThese results suggest that expression of Src is associated with the proliferation ofmicrovascular endothelia in PCNS-DLBCL, unlike that in non-CNS-DLBCL. Theabnormal activation or expression of VEGF-Src pathway may play a key role in thedevelopment and progression of PCNS-DLBCL accompanied with endothelialproliferation. This work may provide a novel target for clinical therapy and study ofPCNS-DLBCL patients.
