Objection:To explore the effect of high glucose on the expression of VEGF inglomerular endothelial cells cultured in vitro and the regulatory role of Alprostadil.Methods:(1) Murine glomerular endothelial cells line were cultured in vitor andwere divided into three groups:Normal glucose group, treated with concentration ofD-glucose(5.6mmol/L);High glucose group, treated with concentration ofD-glucose(30mmol/L) and Mannitol group,treated with mannitol(25mmol/L) plusD-glucose(5.6mmol/L). Stimulated these glomerular endothelial cells0h,24h,48h,72h,96h respectively,detected the protein expression level of VEGF byWestern-blot and immunofluorescence.(2) According to the first step, use the drugAlprostadil to intervent the cells in which time the VEGF protein expression isupregulated and then observe the variety of the protein expression of VEGF.Results:(1) Compared with normal glucose group,in the high glucose group,VEGFprotein expression was significantly upregulated at48h,96h(p<0.05)and there is noobvious change at0h,24h and72h.The immunofluorescence also show thefluorescence intensity of VEGF is higher at48h and96h.(p<0.05)(2) After theintervention of PGE1, the expression is downregulated at48h(p<0.05), but no cleardecrease at96h. And they both increase compared with the normal glucose group. Thefluorescence intensity weaken at48h in the ambient with PGE1(p<0.05) and at96h there is evident vary.Conclusions:(1)The mouse glomerular endothelial cells can express VEGFcultured in vitro and the high glucose updregulate its expression.(2)PGE1candownregulated the overexpression of VEGF.
