Objective The purpose of this experiment is to observe several candidate genes’expression under a hypoxic condition, which are VAMP, LIN-10, RAB-3, andSNG-1, so as to clarify the location of hypoxia injury and to find out thehypoxia-related genes, also to find out whether there is a relationship between theselected gene lin-10and the classical hypoxia-related gene hif-1, and to lay thefoundation of subsequent in-depth study of specific molecular mechanism ofneuron defection under hypoxic condition.Design, time and setting This cell biology experiment was performed at Shen Lab ofBiology Department in Stanford University from September2011to February2012.Materials All the mutant animals, experimental genes, laboratory reagents and laboratoryinstruments involved in this experiment are from Shen Lab of StanfordUniversity.Methods This experiment can be basically divided into two parts. The first part is toscreen hypoxia-related candidate genes by setting an experiment group andcontrol group with a distinct condition of hypoxia and normoxia in which theanimals were incubated separately. The second part is to find out if there is arelationship between the selected gene lin-10and the classical hypoxia-relatedgene hif-1by injecting the LIN-10plasmid into hif-1mutant and then observe theneuron expression under the confocal fluorescence microscopy.Result In the hypoxia-related gene screening experiments, the kyIs235(-) expressed noobvious fluorescence marks on its’ ventral side and dorsal side neuron, while its’corresponding control group kyIs235(+) showed significant fluorescence marksat the same site. There is no significant difference between every otherexperimental group and its’ control group. In the LIN-10and HIF-1correlation study, there is no significant fluorescence marks in experimental groups A, B, Cand E, but there is a constant fluorescence mark in D group.
