ObjectiveTo investigate the expression of Kruppel-Like Factor4(KLF4) in neovascular endothelial cells during the process of wound healing and the effects on endothelial cells proliferation,migration and angiogenesis in vitro.MethodsMaking skin wound model with24rats.Using immunohistochemistry and image analysis technology to observe the KLF4expression in vascular endothelial cells in the wound edge of the trauma model rats at different time points immediately,2,4,6,8,10,12,14d after the trauma; Culture human umbilical vein endothelial cells (HUVECs) in vitro to detect the changes of the HUVEC proliferation,migration and angiogenesis ability using the methods of Cell Counting Kit-8(CCK-8),cells scratches trials, as well as angiogenesis assay in vitro after transfected with KLF4small interfering RNA (siRNA) for48h.Results Compared with the control group, KLF4expression in neovascular endothelial cells. The HUVECs proliferation,migration and form a tubular structure ability were significantly decreased after transfected siRNA down KLF4expression. The absorbance of HUVECs proliferation, the migration distance and tube number were0.948±0.043,206±5.6,12±0.7(P<0.05) respectively after down KLF4expression compared to the control group1.396±0.025,497±6.2,32±0.7.ConclusionThis study demonstrates that KLF4involves in skin wound healing process and plays an important role in the vascular formation (angiogenes is) during cutaneous wound healing.
