Chronic airway inflammation is one of the common risk factors of respiratory diseases,chronic airway inflammation, often there will be a decline in the frequency of cilia beat,making the airway disorders. This study in order to investigate the mechanism of chronicinflammatory airway epithelial cell cilia beat frequency decreased by rat primary airwayepithelial cell cultures were detected by intracellular reactive oxygen species, mitochondrialmembrane potential, LTB4content, and cilia beat frequency indicators. Found in rat airwayepithelial cells in3h by cigarettes and lipopolysaccharide stimulated increase in theconcentration of reactive oxygen species and mitochondrial membrane potential,intracellular LTB4content increased cell cilia beat frequency decreased, there is asignificant difference(p <0.05). Cholinergic inhibitor ipratropium bromide and cigarettestogether with lipopolysaccharide, intracellular content of LTB4decreased cilia beatfrequency rebound, there is a significant difference(p <0.05). In the use of H2O2stimulatecell to produce LTB4amount and cigarettes and lipopolysaccharide stimulated similar, andno significant change(p>0.05). PEG-SOD, catalase and cigarettes and lipopolysaccharidein the common stimulus decreased after the concentration of reactive oxygen species, andLTB4content, there is a significant difference(p <0.05). Thus, chronic inflammation ofairway epithelial cells, cell cilia beat frequency decline is caused by mitochondrial damagewhich can produce ROS caused by cell cholinergic hyperactivity.Objective:The experiment by using cigarettes and lipopolysaccharide stimulate rat airway epithelial cell in primary culture induct the chronic airway inflammation. Investigate the themechanism of cell cilia beat frequency decrease caused by the chronic inflammation.Methods:1. According to the method of Davidson and others to improve the innovative use of ratairway rings paste block method in cultured rat airway epithelial cell.2. High-speed micro-conceived technology records high-speed cilia beat.3. Using ELISAdetection cell supernatant secreted LTB4protein content.4. JC-1probe to examine mitochondrial membrane potential.5. Using DCFHDAdyes to detect cellular ROS.Results:Cigarette and lipopolysaccharide stimulate rat airway epithelial cell3h, theconcentration of reactive oxygen species and mitochondrial membrane potential increase,intracellular LTB4content increase and cell cilia beat frequency decrease, there is asignificant difference (p <0.05). Cholinergic inhibitor ipratropium bromide with cigarettesand lipopolysaccharide together, intracellular content of LTB4decreased and cilia beatfrequency rebound, there is a significant difference (p <0.05). In the use of H2O2stimulatecell to produce LTB4amount.Conclusions:This study successfully replicated the model of airway epithelial cell cilia beat.Elaborated the mechanisms of the mitochondria damage caused by a decline in cilia beatfrequency; By a description of the experimental findings, in airway epithelial cells inchronic inflammation, the decline of the cilia beat frequency was caused by mitochondrialdamage caused by cell cholinergic hyperactivity. lay the theoretical foundation for the studyof cilia beat and related mechanisms.
