Neural connectomics is an emerging discipline, and is to study and to map the brainnerve cell connection (synapse) map. The first important problem which neuralconnectomics need to solve is high brightness and high precision marking of neurons, sothat the imaging device can detect the spines and axon endings. Our group explore theDNA molecular marking method of nerve cells. In order to achieve high brightnessmarking, we construct a variety of series yellow fluorescent protein plasmids; in order toachieve to mark dendritic spines, we construct the fluorescent protein plasmid which hasthe positioning sequence. The author will identify in cultured nerve cells whether theseplasmids can achieve the purposes or not.The fetal rat neural cells in vitro culture method has been established, the authorobtain a variety of yellow fluorescent protein plasmids and the fluorescent protein plasmidwhich has the positioning sequence. The purpose of the subject is to observe theexpression in nerve cells of a variety of fluorescent protein plasmids and the expression innerve cells of the fluorescent protein plasmid which has the positioning sequence. Theexperiment explores whether the brightness of the fluorescent protein is increased,whether fluorescent protein expresses in the nerve fiber. Through reading the literaturesand practicing, and drawing lessons from our laboratory fetal rat neural cells in vitroculture method, I learn the fetal rat neural cells in vitro culture method, culture the neuralcells, and harvest good nerve cells which grow well. Through reading the literatures, Itransfect nerve cells using the liposome and observe fluorescent protein expression usingthe fluorescence microscope. I observed that nerve cells and Hela cells express fluorescentprotein, observed fluorescent protein localizes on the fibrous structures of nerve cellsprotuberances, indicating that a variety of yellow fluorescent protein plasmids and thefluorescent protein plasmid which has the positioning sequence expresses in culturednerve cells, the fluorescent protein plasmid which has the positioning sequence canexpress on the fibrous structures of nerve cells protuberances. I conclude that a variety ofyellow fluorescent protein plasmids and the fluorescent protein plasmid which has the positioning sequence could express in cultured nerve cells, the fluorescent protein plasmidwhich has the positioning sequence could express on the fibrous structures of nerve cellsprotuberances.
