[Background]:Ulcerative colitis(ulcerative colitis,UC),also known as non-specificulcerative colitis,and its prevalence in the home and abroad shows an upward trend. Itsetiology and pathogenesis is not yet entirely clear,clinical treatment consists mainly of5-aminosalicylic acid glucocorticoids,immunosuppressants,etc,but these drugs effect ismore than ideal and has many side effects. Chronic persistent,recurrent is the difficulty ofthe clinical treatment of UC. Erythropoietin (EPO),in addition to used in anemiatreatment related with renal failure and blood system in recent years, the study foundthat EPO has an inhibition of apoptosis,to help vascular repair,enhancements to recoveryand reduce the product of inflammatory factors,to improve humoral and cellular immunefunction,has broad application prospects. It is necessary to explore deeply the EPOtreatment of UC mechanism,also contributes to the pathogenesis of UC research. In thisstudy,using mice UC model and exploring the effect of IL-21/Stat3and Th17cells factor inthe pathogenesis of UC and EPO treatment on IL-21/Stat3and Th17cytokines expressionand its possible mechanism of action.[Objective]:Discussion on the role of Th17in the pathogenesis of ulcerative colitis inmice and observe the possible mechanism of erythropoietin therapy on ulcerative colitis inmice,and which will provide a theoretical basis for EPO bring into clinic in the future.[Method]:Using dextran sodium sulfate-induced mice acute UC model,30BALB/Cmice were randomly divided into5group:normal group,DSS model group,physiologicalsaline(NS)group,erythropoietin(EPO)group,dexamethasone(DXM) group,each group of6.Observing the mouse body weight,stool, diet etc every day,the mice werekilled eight days,the conventional paraffin sections of lesions of the colon,HE staining,calculating the mouse disease activity index score(DAI score),histological damage scoreand the histological pathological changes,using enzyme linked immunosorbent to assayserum interleukin IL-6, IL-17A, IL-21content in mice,immunohistochemical assayingfor the detection of colonic mucosal Stat3expression.[Results]:The symptoms of erythropoietin group and dexamethasone group mice hadmore significantly improved than the model group and the saline group,the score of DAIand histological damage score were low, serum IL-6, IL-17A and IL-21content werelower than the model group and the saline group(P<0.05),the colonic mucosal Stat3expression of erythropoietin group had lower level than the model group and the salinegroup decreased (P<0.05).[Conclusion]:Erythropoietin probably adjusted the cytokine network balance,regulated immune function,controlled intestinal inflammation,and reached the treatmentpurpose of UC finally by decreasing the serum IL-6,IL-17A and IL-21level,reducinglesion intestinal local Stat3expression,blocking IL-21/Stat3anticrime loop and reducingTh17cell factor expression, and it has a considerable effect of dexamethasone.
