At present, scientific researchers for the space environment effects on cellproliferation of studies have focused on the effect of microgravity on cellsproliferation effect. Under the condition of microgravity environment, by inhibitingcell proliferation rate, after microgravity stimulate the cells into the cell cycle speedslows down and the number of cells in G1phase increased obviously and thenumber of cells in S phase and G2phase cuts down. Our previous research workshows that the simulated microgravity effect can make the bone sarcoma cells (U-2OS) of the cell cycle change and make its blockade in G2/M phase. Whether thissuggests we simulated microgravity effect will also be to human peripheral bloodlymphocytes (PBL) affect the cell cycle, so this research intends to consider effectsof simulated microgravity on both PBL from molecular level the influence of cellproliferation.Gene expression profile chip technology is a new technology developed inrecent years and the technology can lead to high flux, high efficient and fast massdetection of gene expression in different samples, its biggest advantage is a changedin the past the traditional experiment can only have one or several gene expressiondifferences were observed. Through the above tips, so this research intends tofurther study simulated microgravity effect on the molecular mechanism of humanperipheral blood lymphocyte cells proliferation. When astronauts into space toefficient work, can maintain health of body and mind is ready for protection.The results show that using flow cytometry instrument to detect simulatedmicrogravity effect72hours make people peripheral blood lymphocytes in G1phaseof the block; Detection of simulated microgravity effect through chromosometechnology72hours of human peripheral blood lymphocyte cells sometimessplitting index found that effects of simulated microgravity cell count of peripheralblood lymphocyte cell division phase decreases; Using the chip technology showedthat effect of simulated microgravity can influence the expression of peripheralblood lymphocyte proliferation related genes CCNA2, CDC2, BUB1, MAD2L1,PCNA, IFI27and KIF2C etc, which also resulted in the changes of the cell cycle.
