Objective: To investigate the expression of Musashi-2and CD133in tissueof colonic adenocarcinoma,and their correlation with the occurrence anddevelopment of colonic adenocarcinoma.Methods: Immunohistochemistry staining was performed to detect theexpression of Musashi-2、CD133、hMLH1and hMSH2in40cases of coloncancer tissues and15cases of normal colonic mucous membrane.Results: The expression of Musashi-2and CD133protein were60%(24/40)and27.5%(11/40)in40colonic adenocarcinoma samplesrespectively.The expressions of Musashi-2and CD133proteins were26.7%(4/15)and0(0/15) in15normal colonic structure samples.There was ahigher expression level of Musashi-2and CD133protein in colonicadenocarcinoma samples than that of normal colonic structure(P <0.05).Therewere significant differences in expressions of Musashi-2proteins betweendifferent histological stages, TNM staging(P <0.05).There were significantdifferences in expressions of CD133proteins between different histologicalstages,lymph node metastasis (P <0.05).But there was no significant differencebetween age and gender.The more poor differentiation and deep infiltratingappear the more lymph node metastasis and distant metastasis, higher TNMstages and higher Musashi-2positive expression were.And the more poor differentiation and the more lymph node metastasis and higherCD133positiveexpression were.The expression of hMLH1and hMSH2protein were62.5%(25/40) and60%(24/40) in40colonic adenocarcinoma samplesrespectively.The expressions of hMLH1and hMSH2proteins were93.3%(14/15)and100%(15/15) in15normal colonic structure samples.There wasa deeper expression level of hMLH1and hMSH2protein in colonicadenocarcinoma samples than that of normal colonic structure(P <0.05).Therewere significant differences in expressions of hMLH1and hMSH2proteinsbetween different histological stages,(P <0.05).Conclusions: Musashi-2and CD133may be related with the initiated anddevelopment of colonic cancer. They may become a new auxiliary clinicaldiagnosis and therapeutic targets. The loss or reduction of mismatch repair genehMLH1and hMSH2are early events in tumorigenesis, possibly associated withadenocarcinoma of colon, it is significant to study on the occurrence of colonadenocarcinoma。...
