Study On The Secondary Metabolites Of Three Marine Organisms And Their Antibacterial Activity

In order to obtain new bioactive lead-compounds from marine organisms as the material base for efficient, safe and nontoxic natural food preservative, the chemical components of gorgonian Menella sp., Subergorgia reticulata, and bacterium were investigated systematically, and the antimicrobial activity of some of the compounds isolated from Menella sp. and Bacillus subtilis were also evaluated.19compounds were isolated and purified from both of the the ethyl acetate and n-butanol extracts of Menella sp. by the methods of CC, Sem i-preparative HPLC, TLC, Sephadex LH-20and so on. By comparison wit h literatures and on the basis of modern spectroscopic analysis, the struct ures of15compounds among the19compounds were identified as1,2-B enzenedicarboxylic acid,1,2-bis(2-methylpropyl) ester (M3),2-methylimida zoline (M4), Nicotinamide (M5), Cyclo(L-leucyl-trans-4-hydroxy-L-proline)(M6),3-Methyl uracil (M7), Thymine (M8),7-Methyladenine (M9), Hypo-xanthine (M10), Uracil (M11), Uracil deoxyriboside (M12), Adenine deox-yribonucleoside (M13), Thymine (M14), Thymine deoxyribonucleoside (M15). Two compounds (M1, M2) of them were new.8compounds were isolated and purified from the ethyl acetate extra-cts of gorgonian Subergorgia reticulata. By comparison with literatures and on the basis of modern spectroscopic analysis and the physicochemica1properties as well, the structures of the8compounds were identified as cholest-5-en-3β-o (S1), Uracil (S2), Thymine(S3), Nicotinamide (S4),1,3,9-trimethyl-lH-purine-6,8(7H,9H)-dione (S5), Caffe-ine (S6), Adenine (S7), p-Hydroxybenzonic acid (S8). Compounds S1, S5, S6, S7, and S8we-re found in this animal for the first time.Study on the secondary metabolites of gorgonian-associated bacteria Bacillus subtilis.11compounds were isolated from the ethyl acetate extra-ct of the fluid medium of Bacillus subtilis. By comparison with literatures and on the basis of modern spectroscopic analysis,7compounds among the11compounds were identified as2-O-butyl-1-O-(2’-ethylhexyl)benzene-1,8-dicarboxylate (B2), DEHP (B3), Cyclo(proline-leucine)(B4),2-(hexade-canoylamino)-4,8-Octadeca-diene-1,3-diol (B5), C14-Sphingosine (B6),2-(4-Aminophenyl)ethanol (B7). One of them was a new Compound (B1)Eleven compounds(Ml-M3、M6-M9and B1-B4) were evaluated for their bacteriostatic activity. M1,M2and B1showed remarkable activities against four of food spoilage organisms, which can be used as natural food preservatives in potential.Moreover, terpenes from gorgonian in recently thirty years and the secondary metabolites of gorgonian-associated bacteria were summarized in the first chapter of the thesis.
