In this study, imported from Canada transgenic chlorella vulgaris as raw material, to determine the appropriate medium to explore the factors that affect to turn of transgenic chlorella vulgaris growth, optimize the transgenic chlorella vulgaris optimal culture conditions, and To optimize the extraction process of transgenic chlorella vulgaris fatty acids,, the results are as follows:1. Analysis examines to determine the experimental algae transgenic chlorella vulgaris, media GMSC medium.Take the single factor experiment transgenic chlorella vulgaris culture conditions to optimize, identified the most transgenic Chlorella vulgaris cultivation conditions, the results are as follows:Ratio of Urea and KH2PO4:4:3PH:8.5Culture temperature:28℃Cultured under this condition transgenic Chlorella vulgaris extract of total fatty acids, the results show that the fatty acid yield can reach21.94%.2. Take the single factor experiments to optimize the process of extracting fatty acids from transgenic chlorella vulgaris, to determine the optimal fatty acid extraction process, and the results are as follows:Extraction Method:chloroform-methanol extractionThe amount of solvent:1g dry algae powder add10ml of methanol and20ml of chloroformExtraction times:twoExtraction temperature:40℃Under the conditions of the above optimization the transgenic chlorella vulgaris extract, fatty acid extraction was18.10%. Wherein Linoleic acid content of7.734%of the total fats, and the linolenic acid content of15.544%of the total fat,13.239%of the total fat content of oleic acid, stearic acid content of1.442%of the total fat content of palmitic acid17.766%of the total fat.The experimental optimization of culture conditions transgenic chlorella vulgaris and fatty acid extraction process, provide a theoretical basis for the transgenic chlorella vulgaris large-scale high-density culture and fatty acid industrial production, transgenic chlorella vulgaris production of fatty acids in the technical support.
