Pachymaran, which is extracted form poria cocos, has a wide variety of biologic-al acivities, such as enhancing body immuneity, antitumor, sedation, diuresis, detume-scence and so on. However, pachymaran is poor in water-soluble, therefore, the effort of chemical modification in structure could be took on it, and has significance to incr-ease water-soluble, reveal relationship of structure-activity, and improve the use of pa-chymaran.Experiment discussed the influence factors of method of ultrasonic aided extract-ion alkali soluble pachymaran. Through the chemically modify of acylation, sulfonati-on and esterification, molecular structure of pachymaran was changed, and the struct-ure of derivatives were characterized. Single factor and orthogonal experiment were carried on to compare the influence factors of preparative reaction of acetylated-pach-maran and determine the optimum reaction conditions. The main results obtained thr-ough experiments were as follows:Room temperature, ultrasonic aided extraction alkali soluble pachymaran on the optimum technological conditions:solid-liquid ratio1:50(g/mL), concentration of NaOH0.75mol/L, extracting time60min, power96w, the yield of alkali soluble pa-chymaran was87.53%.With dimethyl sulfoxide solvent, acetic anhydride as acetylating agent, N-bromo-butanimide (NBS) as catalyst, acetylated-pachymaran was prepared. Orthogonal exp-eriment wes carried on to determine the optimum process of preparation. Results indi-cated that the preparation of acetylated-pachymaran on the optimum technological co-nditions for:acetic anhydride/pachymaran, the mass ratio of chitosan volume of12.5:1(mL/g), NBS/pachymaran, the mass ratio of chitosan0.125:1(g/g), reaction temperature80℃, reaction time5h.Sulphonating reagent was prepared by triethylamine and benzene sulfochloride in ice salt bath condition, then respectively to dimethyl sulfoxide, N, N-DMF, THF as solvent, on to the modify of sulphonated pachymaran. Results indicated that in di-methyl sulfoxide as solvent, sulphonated pachymaran was able to acquire high yield.With ferulic acid for esterification reagent, dimethyl sulfoxide as solvent, N, N-DMF as solvent, benzene, toluene as silane, ferulate of pachymaran was prepared, the physical and chemical properties were analysized, and structure characterization was identificed.The antioxidation activities of polysaccharide derivatives on hydroxyl radicals (·OH), oxygen radicals(·O2-) and DPPH·free radicals were tested. Results indicated t-hat the polysaccharide derivatives prepared by pachymaran have reducing power can detecting hydroxyl radical-s(·OH), oxygen radicals(·O2-) and DPPH·free radicals dee-ply.
