Rabies has a very high rate of death. Once a person or animal infected withRV, The RV can spread with blood from the site of infection to the brain. In theroyalthough all animal and humanity can be communicator of RV, but, in fact, the mostof RV is communicated by dog, cat and cat. In present, people can not still curerabies,but controlling and preventing is possible.In our country, the RV epidemic is very serious. In the world, india occupy thefirst in the epidemic, the second is the China. In the21st century, with the petsincreasing, there is an increasing tend. Our goverment take two measures against RVepidemic. One is called killing that means kill animal that can spread. The other isthat make use of vaacine. In our government, main RV vaccine is injection vaccine.Injection vaccine have some shortcoming. For example, the immuning process iscost and complex. And sometimes, some workers are bited and harmed. What,s more,It can not be widely used to immune wild animal. The oral vaccine can overcmoethese shortcoings. It is possible that make use of oral vaccine to immune all the wildanimal.Objective: In this study, we take Flury strain and alginate sodium as anexprimental model to prepare calcium alginate gel beads of oral live attenuatedrabies vaccine. The vaccine product was studied in terms of the preparation, theimmune results and sterility.Means: In the first, the combination between a single-factor test and orthogonaltest to optimize the preparation of AGS gel beads.Then vaccine product wasobtained by the best preparation. The third, the drug release is studied by “therotating basket”. The forth, different culture was made up to check the sterility oforal vaccine. The fifth, the cell toxity test in vitro was carried out by MTT. The toxitytest in vivo was studied by the means commended by WHO. Finally, we chosednomal Kunming mouse as experimental animal.The animal expriment was studies to check the immunity. cellular and humoralimmunity was respectively studied by the FAS and the MNT.Result: by optimizing the preparation, the best preparation was found. The finalpriduct is obtained by the best preparation that loading effective was17.49±1.43%and the entrap effiency was78.64±2.144%. In the sterility test, The different kinds of cultures is found that there is no bacterium.The cell toxity testshowed that the product had no cell toxity after120h that product interact with L929cell. Toxity test in vivo proved that the final product have no toxity in vivo with theexprimental animal growing healthly.In the final animal experiment,immuned animalhave better the effect of cellular and humoral immunity. In the cellular immunity, it isconcluded that the number of CD4+cells had an apparent rise from25.81±3.21%to41.91±2.32%, with the increased number of CD8+cells from20.51±2.26%to25.70±2.11%. There was also a rise of (?)/(?) from1.26±0.31to1.63±0.48that proved the body immunity have improvement.Conclusion: As a expriment model of a comination of Flury strain and alciumalginate sodium as a attenuated oral rabies vaccine, attenuated oral rabies vaccinewas preliminarly studied, and and got quite a better immune effect. The studyprovides a safe, easy-to-use and low-cost path to RV vaccive.
