Monosodium glutamate (MSG) was often used as a flavor additive in food. But it is a controversial issua that whether safe about dieting MSG or not. On the one hand, its main ingredient is sodium glutamate which is decomposed into glutamic acid as important physiological role in the gastrointestinal tract. On the other hand, there may be damaged body. Many reports identified the MSG is known as the culprit of "Chinese Restaurant Syndrome". The objective of the present study was to observe effects of glutamate on physiological metabolism and nutrition health of ordinary female, pregnancy maternal and its progeny by adding MSG (1%) in mixed feeds.Part Ⅰ:Lee’s index (P<0.05) and BMI (P<0.01) of female rats were been effected by MSG (1%), and there were significant differences for the heart index and kidney index (P<0.05). MSG (1%) significantly increased the number of the white blood cells, the number of lymphocytes, monocytes, neutrophils, and the total number of red blood cells compared with control group. There were significant increase of the number of ALP, CR, UI, serum free amino acids, essential amino acids and non-essential amino acid. MSG (1%) affected Lee’s index (P<0.05) and BMI (P<0.01) of lactating rats, too. But there were little effect on the number of white blood cells, lymphocytes, monocytes, neutrophils, and the total number of red blood cells, MSG (1%) just increased the number of platelets. Compare with control group, the number of ALP, CR, UI, serum free amino acids were significantly increased in experience group, but total amino, essential amino acids and non-essential amino acid were lower trend in experience group.Part Ⅱ:Liver cell morphology of MSG (1%) female rats group was polygonal, but less regulation, the liver plate arranged slightly messy, fat particles more than control group and there were many inflammatory cells. Liver cell morphology of1%MSG lactating rat group were polygonal, but they were less regulation, too. The liver plate arranged slightly messy, there were swelling liver cells around the central vein, sinusoidal wasn’t uniform, glycogen and fat particles more than control group’s and there were many inflammatory cells. The results indicated that MSG (1%) damaged liver. Glomerulus of kidney were hypertrophy both female and lactating group with1%MSG. Bowman’s capsule were more thicker, endothelial cells arranged in regular and had an exfoliation phenomenon. Glomerulus of MSG (1%) lactating rat group were normal morphology. Bowman’s capsule were more thicker, endothelial cells arranged less regulation and had an exfoliation phenomenon. The results indicated that1%MSG damaged kidney.MSG (1%) experimental rat group, the duodenum, jejunum, ileum and colon mucosa, complete fluff arranged in neat rows, goblet cells were clear and obvious, and a small amount of lymphocytes were existed. MSG (1%) group villi were longer and more close arranged compared with control group. Colonic mucosa glands were evenly, goblet cells were seen clearly in female experimental group and lactaing experimental group, and there were some lymphocytes in glands. Compared with control group, mucosal were more thicker in experimental group. The results indicated that MSG (1%) can promote the growth of intestinal villus.Part Ⅲ:Offprings were observed, there were significant difference for Lee’s index and BMI index, which indicated that MSG (1%) led to central obesity of offspring. In the neurobehavioral experiment, there were significant differences between experimental group and control group. The results indicated MSG (1%) effect sensorimotor ability of offspring. In the open field test, the number of across grid and standing in experimental group were lower than control group’s, while the number of bowel movements and the frequecy of stroking hair were significantly higher. The results showed that MSG (1%) diminished the self-exploration and the movement capacities of the offspring, which accompanied with anxiety. There were not significant difference in Morris water maze test in experimental group compare with control group. The results showed that MSG (1%) had no effect on learning and memory of offspring.
