Objective:This study selected the protein serpinA3and PEBP1which were root in recentproteomics of gastric cancer, detected their expression in different gastric tissues andanalyzed the clinical significance, provides some experimental basis for earlydiagnosis of gastric cancer.Methods:Collected normal gastric mucosa, gastric cancer and paracancerous tissues;picked up total protein and RNA in different tissues; then detected the expression ofserpinA3and PEBP1protein in different gastric tissues by using Western-blot andRT-PCR. At the same time, detected the expression of serpinA3and PEBP1protein oncertain tissue microarray (normal gastric mucosa,atypical hyerplasia and gastriccancer) by using the immunohistochemistry staining. In the end, analysis therelationship between their expression and the development of gastric cancer as well asthe clinical pathology significance.Results:1. Western-blot showed that the level of expression of serpinA3protein washigher in gastric cancer and gastric carcinoma tissues than in normal gastric mucosatissue (P<0.01); The expression of PEBP1in gastric cancer tissue was up-regulatedcompared with normal gastric mucosa tissues, but there were no obvious difference inadjacent tissues and normal gastric mucosa tissues.2. RT-PCR showed the expression level of the serpinA3and PEBP1representedgradually decreased in gastric cancer, gastric carcinoma tissues and normal gastricmucosa tissues. But the diversity between gastric carcinoma tissues and normalgastric mucosa was unobvious. 3. The immunohistochemical staining consequence exhibited that the expression rate of serpinA3protein in normal gastric mucosa,atypical hyerplasia, gastric cancer and gastric cancer tissues with lymph node metastasis was4.16%(1/24)、18.75%(3/16)、62.96%(34/54)and73.52%(25/34),The positive expression in well-differentiated,moderately differentiated and poorly differentiated gastric cancerwere50.00%(3/6)64.28%(9/14)and79.41%(27/34); The positive expression rate of Pebp1protein in normal gastric mucosa,atypical hyerplasia, gastriccancer and gastric cancer tissues with lymph node metastasis was5.00%(1/20),22.22%(4/20),70.37%(31/52)and66.66%(22/33),The expression rate in well-differentiated,moderately differentiated and poorly differentiated gastric cancer were60.00%(3/5),66.66%(6/9)and81.57%(31/38).The positive expression rate of serpinA3and pebp1protein in cancer tissue were higher than the normal gastricmucosa and atypical hyperplasia tissues (P<0.01); Their expressed rate in thelymph node metastasis cancer were increase compare with in the primary carcinoma(P<0.05).Conclusions:1. The expression level of serpinA3and Pebp1protein in gastric cancerwas up-regulated compare with atypical hyerplasia tissue and normal gastricmucosa tissue,their positive expression rate in the gastric cancer with lymphnode metastasis were higher than in the primary carcinoma.2. The high expression of serpinA3and Pebp1protein may be relatedto the development and metastasis of gastric cancer.
