Objective To detect role effect of using huaier particles on the different expressionof estrogen receptor of human breast cancer cells, and study its possible mechanism.Methods To cultivate MCF-7(ER+) and MDA-MB-231(ER-) cells in vitro,and usethe role of different concentration of huaier particles (1mg/ml、2mg/ml、4mg/ml、8mg/ml) treat them.The inhibitory effect of huaier particles on the MCF-7andMDA-MB-231cell was investigated with3-(4,5-dimethyl-thizazol-2-yl)-2,5-diphenylbromide(MTT). Inverted microscope observe cell morphology; Cell cycle wasmeasured by flow cytometry; Apoptosis was detected with Annexin V-FITC+PI dualparameter.Results (1) MTT assay show that huaier particles could inhibit the growth of thehuman breast cancer cells MCF-7and MDA-MB-231; With the increase of drugconcentration and action time, inhibition enhanced gradually, and the inhibitory effectwas dose and time-dependent manner(P<0.05).When the concentration was8mg/mlfor72h, the MCF-7cell inhibition rate was80.10%;and the MDA-MB-231cellinhibition rate of76.42%;but when the same drug concentration and phase, Inhibitionrate of two kinds of cells were no difference statistically.(2) MCF-7and MDA-MB-231cells morphology variation with concentration wereobserved under the inverted microscope. Cells in the control group (not treated withhuaier particles), MCF-7cell was polygonal in shape, MDA-MB-231cell was ovoidor spindle, and the two kinds of cell lines were intercellular tight junction and vaguelyoutline, had transparent and homogeneous cytoplasm, and were growing with goodcondition. With the increase of huaier particles concentration, Cells were out ofcondition, adherence cells decreased, cells became round and small, Particulate matter increased and cells became round and small. Floating cells were in creased in theculture flask gradually.(3) Huaier particles can make MCF-7and MDA-MB-231cells significant changes incell cycle. The cells in G1stage multed as the density of concentrations increased, Sand G2stage ’s cells decreased gradually; compared with the control group, thedifference was statistically significant (P <0.05). As huaier particle treatment cells in72h, MCF-7cells in G1stage of the experimental group accounted for69.82%of thetotal number of cells, and MDA-MB-231cells in G1phase reached for71.79%.(4) Streaming Annexin V/PI double staining result show that:the early apoptosis rateof MCF-7and MDA-MB-231cells have significant difference(P<0.05); With theincrease of drug concentration and action time,apoptosis rate increased obviously, theapoptosis rate was dose and time-dependent manner(P<0.05);but when the same drugconcentration and phase, two kinds of cells were no difference statistically.Conclusions (1) Huaier particles have value-added inhibition and induction ofapoptosis on MCF-7and MDA-MB-231cells in vitro;and there is a time dose-effectrelationship.(2) Huaier particles can inhibit both cell cycle at G1phase.
