Objective:To investigate the effect of high glucose on the production of NO,TNF-α andICAM-1in cultured human umbilical vein endothelial cells (HUVECs), and to furtherstudy the intervention of polysaccharides of dendrobium candidum (PDC).Methods:The cultured HUVECs in vitro were as the object to investigate the protectiveeffects of polysaccharides of dendrobium candidum on high glucose-inducedinflammatory injury.They were treated to the logarithmic growth phase, HUVECswere treated with high glucose (33.3mmol/L) for48h to induce the injury.ThenHUVECs were treated with PDC (100,200,400and800μg/mL) for48h. The cellviability was detected by MTT assay;HUVECs were cultured in DMEM mediumwith or without high glucose for different times(24h,48h and72h).The cells weredivided randomly into the control group, control group+PDC (400μg/mL)group,high glucose group and high glucose+PDC (400μg/mL) group. Production ofNO was detected by Griess assay.Meanwhile TNF-α and ICAM-1protein expressionin cells were measured by Western blot(W.B).Results:①Compared with the control group, PDC (100,200,400and800μg/mL) didnot affect the cells viability and apoptosis of HUVECs (all P>0.05).②Compared with the high glucose group, the cell viability of PDC (100,200,400and800μg/mL) was significantly increased in concentration dependent manner(all P<0.05);but compared with PDC (400μg/mL) group,the cell viability of the PDC(800μg/mL) group was not significantly changed(P>0.05).③Compared with the control group, the concentration of NO was significantly increased(P<0.05) at24h, and significantly decreased(P<0.05) at48h in high glucosegroup, and as cultured time lengthened to72h in high glucose group, there was noremarkable change(P>0.05) between72h and48h; Compared with the high glucosegroup, the concentration of NO was significantly decreased(P<0.05) at24h, andsignificantly increased(P<0.05) at48h in high glucose+PDC (400μg/mL) group, andas cultured time lengthened to72h, there was no remarkable change(P>0.05) between72h and48h. Compared with the control group, the concentration of NO was nosignificantly increased(P>0.05) as cultured time lengthened in the control group+PDC (400μg/mL).④HUVECs were treated with PDC (400μg/mL) for48h. Compared with thecontrol group,level of TNF-α and ICAM-1was significantly increased in high glucosegroup(all P<0.05); Compared with the high glucose group, level of TNF-α andICAM-1was significantly decreased(P<0.05) in high glucose+PDC (400μg/mL)group.And there was no difference(P>0.05) between the control group and the controlgroup+PDC (400μg/mL).Conclusions:Polysaccharides of dendrobium candidum inhibits the inflammatory injury byhigh glucose in cultured human umbilical vein endothelial cells,and increase the theconcentration of NO,decrease the level of TNF-α and ICAM-1.
