| ObjectiveThrough observing the effect of coffee acid phenethyl ester (caffeic acid phenethylester, CAPE) on human colon cancer Lovo cells PI3K/AKT signaling pathway,discuss the mechanism of CAPE on human colon cancer Lovo cells.Method1. With concentration respectively0mu g/mL,2.5mu g/mL,5.0mu g/mL,7.5mu g/mL,10.0mu g/mL of CAPE deal with human colon cancer Lovo cells, usingdetermined by MTT method (4methyl azo azole salt) observed at different times (24h,48h,72h) on Lovo cells growth inhibition rate;2. Application of PT-PCR detection were treated with different concentrationsof CAPE Lovo cells after72h of PI3K in the expression of mRNA;3. Using Western blot method analysis obtained with different concentrations ofCAPE Lovo cells after72h of PI3K, and AKT, p-AKT, caspase3, the influence ofcaspase-9protein expression.Result1.10μ g/mL CAPE72h on Lovo cell inhibition rate was61.28%, significantlyhigher than that of2.5μ g/mL72h after29.63%inhibition rate (P<0.05), wassignificantly higher than that of10μ g/mL24h after37.84%inhibition rate (P<0.05);2(2.5~10)μg/mL CAPE on Lovo cell growth were inhibited, presentationtime and dose dependent; 3.PI3KmRNA and its protein expression with elevated CAPE concentration andreduction, the experimental group and the control group had significant difference(P<0.05);4.The expression of AKT protein is not as elevated CAPE concentration changes,the experimental group compared with the control group, no significant difference(P>0.05), the expression of p-AKT protein with elevated CAPE concentration andreduction, the experimental group and the control group had significant difference(P<0.05), Caspase-3, caspase-9protein expression with CAPE concentrationincrease, each experimental group compared with the control group, there wassignificant difference (P<0.05).Conclusion1CAPE can inhibit in vitro human colon cancer Lovo cells proliferation, and ina certain range of concentration and time dependent;2. AKT phosphorylation mechanism of CAPE inhibits the proliferation of humancolon cancer cells may inhibit the PI3K/AKT signaling pathway and CAPE related. |
