Objective: To research the effect of Curcumin Trinicotinate (CurTn) on theproliferation of VSMC in vitro via Caveolin-1/ERK1/2/CyclinD1signaling pathway.Methods: The VSMC were cultured and maintained in DMEM supplemented in vitro,the proliferation of VSMC were induced by10%FBS. MTT assay was used to detectthe effect of CurTn on the proliferation of VSMC. FCM was used to observe theeffect of CurTn on the distribution of cell cycle. Western blotting was applied toobserve the expression of PCNA、Caveolin-1、p-ERK1/2and CyclinD1, and the effectof CurTn on the expression of p-ERK1/2after Caveolin-1silenced with si-RNA ofCaveolin-1and the effect of CurTn on the expression of CyclinD1after VSMCpre-incubated PD98059.Results:MTT assay showed that CurTn inhibited VSMC proliferation induced by10%FBS in time and dose-dependent manner. FCM indicated that CurTn acceleratedtransformation of cell cycle G1/S phase, increased the distribution proportion of cellcycle G0/G1phase and decreased the percentage of cell cycle S phase. Westernblotting displayed that CurTn up-regulated the expression of Caveolin-1protein andsuppressed the expression of p-ERK1/2、CyclinD1and PCNA protein; and si-RNA ofCaveolin-1lowered the effect that CurTn suppressed the expression of p-ERK1/2protein and PD98059lowered the effect that CurTn suppressed the expression ofCyclinD1protein.Conclusions: CurTn will up-regulate the expression of Caveolin-1protein, lead to thedown-regulation of ERK1/2protein expression and cause the reduction of CyclinD1protein expression, accelerate transformation of cell cycle G1/S phase that result cellcycle arrest G0/G1phase, thus inhibiting VSMC proliferation.
