Objective: To detect the expression levels of human epidermal growth factor receptor2(HER-2) and growth hormone(GH)/growth hormone receptor(GHR) in the serum andtumor tissue of gastric carcinoma,and investigate their function in the genesis anddevelopment of gastric carcinoma, and then provide the theory basis for the diagnosis,prognosis evaluation and guide therapy of the patients.Methods:1. The preoperative serum and postoperative tumorous tissues of84patients with gastriccarcinoma served as serous and tissular experimental group, respectively, the serum of20normal persons as serous control group,and the postoperative corresponding para-tumorous tissues of84patients with gastric carcinoma as tissular control group,enzyme-linked immunosorbent assay (ELISA) and immunohistochemistry (IHC)methods were respectively used to detect the expressions of HER-2and GH/GHR in theserum and tissue.2. The relationship between the expressions of HER-2and GH/GHR and the genesisand development of gastric carcinoma were analyzed.3. All data were processed by SPSS13.0.Results:1. Expression levels of serum HER-2The average levels of HER-2of the serous experimental group(13.21±6.69μg/L)was obviously higher than that of the serouscontrol group(5.40±1.04μg/L), there was statistically significant between the two(P<0.05). The positive rate of HER-2of the serous experimental group(20.2%)wasobviously higher than that of the serous control group (0), there was statisticallysignificant between the two(P<0.05).Serum HER-2levels was not correlated withgender,age,tumor site,tumor size, differentiation, depth of invasion, lymphaticmetastasis, distant metastasis, TNM stage and Lauren classification(all P>0.05);2. Expression levels of tissue HER-2The positive rate of HER-2of the tissularexperimental group(29.8%)was obviously higher than that of the tissular control group(0), there was statistically significant between the two(P<0.05).The high tissueHER-2levels was significant correlation with the better differentiation, high TNM stage,lymphatic metastasis and Lauren classification(P<0.05),but not with gender,age,tumorsite,tumor size, depth of invasion, distant metastasis(all P>0.05);3. Correlation between the expressions of serum HER-2and tissue HER-2ofgastric carcinoma The expression levels of serum HER-2was positively correlatedwith tissue HER-2in patients of gastric carninoma(χ2=8.611,r=0.320,P=0.003);4. Expression levels of serum GH The average levels of GH of the serousexperimental group(6.00±2.95μg/L)was obviously higher than that of the serouscontrol group(0.78±1.15μg/L), there was statistically significant between thetwo(P<0.05). The positive rate of GH of the serous experimental group (20.2%) wasobviously higher than that of the serous control group (0), there was statisticallysignificant between the two (P<0.05). The high serum GH levels was significantcorrelation with the large size and lymphatic metastasis(P<0.05), but not withgender,age,tumor site, differentiation, depth of invasion, distant metastasis, TNM stageand Lauren classification(all P>0.05);5. Expression levels of tissue GHR The positive rate of GHR of the tissularexperimental group (56.0%) was obviously lower than that of the tissular control group(84.5%), there was statistically significant between the two(P<0.05). The high tissue GHR levels was significant correlation with the better differentiation(P<0.05),but notwith gender,age,tumor site,tumor size, depth of invasion, lymphatic metastasis, distantmetastasis, TNM stage and Lauren classification(all P>0.05);6. Correlation between the expressions of serum GH and tissue GHR of gastriccarcinoma The expression levels of serum GH was negatively correlated with tissueGHR in patients of gastric carcinoma(χ2=11.919,r=-0.405,P=0.001);7. Correlation between the expressions of tissue GHR and tissue HER-2of gastriccarcinoma The expression levels of tissue GHR was positively correlated with tissueHER-2in patients of gastric carcinoma(χ2=8.352,r=0.315,P=0.003)。Conclusion:1.The high levels of serum HER-2and tissue HER-2is significantly correlated with thegenesis and development of gastric carcinoma, and which can provide the theory basisfor the diagnosis, prognosis evaluation and targeted therapy of the patients;2. The expression levels of serum HER-2was positively correlated with tissue HER-2inpatients of gastric carninoma,and thus detecting the serum levels of HER-2can reflectthe tissue levels of HER-2of the patients;3. The high levels of serum GH and the low levels of GHR is significantly correlatedwith the genesis and development of gastric carcinoma, and which can provide thetheory basis for the diagnosis, prognosis evaluation and endocrine therapy of thepatients;4. The expression levels of serum GH was negatively correlated with tissue GHR inpatients of gastric carninoma,and thus detecting the serum levels of GH can reflect thetissue levels of GHR of the patients;5. The co-expression pattern of HER-2and GH/GHR in patients with gastric carcinomasuggest that they may act synergistically in the genesis and development of gastriccarcinoma.
