Background Psoriasis (OMIM*177900) is a chronic, common, recurrent andinflammatory skin disease with different prevalences among viriaty of populationsworldwide. The pathogenesis of psoriasis remains unclear. Genetic factors, immunologyand environmental factors play important roles in the pathogenesis of psoriasis. In total,13susceptibility loci (PSORS1-13) have been successfully identified. Some studiesprovide evindence psoriasis share susceptibility genes with other disorders (e.g.Ankylosing spondylitis, Crohn’s Disease). About10%of AS cases have psoriasis, andboth are associated with the IL23R and ERAP1genes. The SNPs rs4672495(P=3.2×109) at2p15, rs4333130(P=9.3×108) within ANTXR2at4q21and rs2242944(P=8.3×1020) at21q22are associated with AS. rs4552569(P=8.77×1010) betweenEDIL3and HAPLN1at5q14.3and non-synonymous single nucleotide polymorphism(nsSNP), rs4077515in CARD9(caspase recruitment domain-containing protein9) on9q34.3, rs2297909in KIF21B at1q32as well, are shown association with AS inChinese Han and European populations, although there is genetic heterogeneity.Objective To investigate the potential association of8SNPs with psoriasis in ChineseHan, we performed a large scale replication study (2208cases and2208controls) basedon the result of AS in the European population.Methods We replicated these8SNPs in Chinese Han using SequenomMassarraysystem and analysized the date by Plink1.07statistically.Results1. The distributions of alleles and genotypes for8SNPs are not differentbetween cases and controls(P>0.05).2. All cases were stratified by clinical types, thereis difference for rs2297909among family history and age of onset subgroups ofpsoriasis (P <0.05)。3. There is difference for rs6854711between controls and onset age of psoriasis (P <0.05).4. No difference is found in distributions of alleles andgenotypes for the other SNPs between clinical subgroups (P>0.05).Conclusions rs2297909is associated with family history and age of onset for psoriasisin Chinese Han; rs6854711is associated with age of onset for psoriasis in Chinese Han.The other SNPs show no association with psoriasis, maybe resulting from geneticheterogeneity in different populations in these two diseases.
