Objective This study aimed to investigate the expression and clinical signification ofCD4~+CD25~+Tregs,Foxp3,TGF-β1in peripheral blood and cancerous tissue of gastriccancer.and investigate their function in the genesis and development of gastriccarcinoma in order to comprehend its value in clinical aspect which contribute to theearly diagnosis and immunotherapy of gastric cancer.Methods1.Object of study:To collect preoperative blood sample,gastric cancer tissue andpara-tumorous tissue,normal tissue of38patients with gastric cancer in the FirstAffiliated Hospital of Anhui Medical University from April,2012to November,2011,which are gastric cancer group,and blood sample of20cases of healthy control group inthe hospital health examination center.We divided patients into different groups whichare TNM stage,differentiation,lymph node metastases,tumor site and tumor diameter indifferent ways according to the needs of data analysis.2.FCM was applied for gastric cancer patients and healthy control group in theperipheral blood to test CD4~+CD25~+Foxp3~+Tregs expression.Those from gastric cancerpatients were made analysis with different groups which are TNM stage,differentiation,lymph node metastases,tumor site and tumor diameter. 3.IHC was applied for gastric cancer patients in gastric cancer tissue and para-tumoroustissue,normal tissue to test CD4~+CD25~+Foxp3~+Tregs expression.Those from gastriccancer tissue were made analysis with different groups which are TNM stage,differentiation,lymph node metastases,tumor site and tumor diameter,and correlationanalysis with CD4~+CD25~+Foxp3~+Tregs expression in the peripheral blood.4.ELISA was applied for gastric cancer patients and healthy control group in the serumto test TGF-β1expression.Those from gastric cancer patients were made analysis withdifferent groups which are TNM stage,differentiation,lymph node metastases,tumor siteand tumor diameter,and correlation analysis with CD4~+CD25~+Foxp3~+Tregs expressionin the peripheral blood.Results1.The expression of CD4~+CD25~+Foxp3~+Tregs in the peripheral blood: The expression ofCD4~+CD25~+Foxp3~+Tregs in peripheral blood from gastric cancer group weresignificantly higher than those from healthy control group(P<0.001),and were highlycorrelated to TNM stage,lymph node metastase(sP<0.001,P<0.001).The expressionof CD4~+CD25~+Foxp3~+Tregs in the TNM stage were compared between Ⅰ~+Ⅱ stage, Ⅲstage and Ⅳ stage each other were significant(P=0.001,P<0.001,P<0.001).2.The expression of CD4~+CD25~+Foxp3~+Tregs in gastric cancer tissue:The expression ofCD4~+CD25~+Foxp3~+Tregs in gastric cancer tissue,para-tumorous tissue,normal tissuewere significant difference (P<0.001),and those in gastric cancer tissue weresignificant than those in para-tumorous tissue,normal tissue(P<0.001,P<0.001).Itwere highly correlated to TNM stage,tumor differentiation,lymph node metastases(P<0.001,P=0.015,P<0.001).Those were compared between Ⅰ~+Ⅱ stage, Ⅲ stageand Ⅳ stage each other were significant(P=0.020,P<0.001,P=0.001)in the TNM stage.Those in the tumor differentiation were compared between well-differentiated,moderately differentiated, poorly differentiated eath other,and those in well-differentiated group were significant than those in moderately differentiated,poorlydifferentiated(P=0.026,P=0.004).The expression of CD4~+CD25~+Foxp3~+Tregs inPeripheral blood was positively correlated to CD4~+CD25~+Foxp3~+Tregs in gastric cancertissue(r=0.786,P<0.001).3.The expression of TGF-β1in the serum:The expression of TGF-β1in serum fromgastric cancer group were significantly higher than those from healthy control group(P<0.001),and were highly correlated to TNM stage,tumor differentiation,lymph nodemetastases(P<0.001,P=0.015,P<0.001).Those in the TNM stage were comparedbetween Ⅰ~+Ⅱ stage, Ⅲ stage and Ⅳ stage eath other,and those in Ⅰ~+Ⅱ stage weresignificant than those in Ⅲ stage, Ⅳ stage(P<0.001,P<0.001).Those in the tumordifferentiation were compared between well-differentiated,moderately differentiated,poorly differentiated eath other,and those in well-differentiated group were significantthan those in poorly differentiated (P=0.004).The expression of CD4~+CD25~+Foxp3~+Tregs in Peripheral blood was positively correlated to TGF-β1in serum(r=0.793,P<0.001).Conclusion1.The expression of CD4~+CD25~+Foxp3~+Tregs is significantly correlated with thegenesis and development of gastric carcinoma,and which can provide the theory basisfor the early diagnosis, prognosis evaluation and immunotherapy of gastric cancer.2.The expression of TGF-β1is significantly correlated with the genesis anddevelopment of gastric carcinoma,and which can provide the theory basis for evaluationTNM stage roughly and immunotherapy of gastric cancer.. 3.The pathogenesis of gastric carcinoma is influenced both by CD4~+CD25~+Foxp3~+Tregsand TGF-β1.
